Background. Immunohistochemical assessment of breast cancer and stratification into the basic molecular subtypes afford a much deeper insight into the biology of breast cancer, while presenting with opportunities to exploit personalized, targeted treatment. Traditionally, the oestrogen, progesterone, and epidermal growth factor receptors are assessed. MUC1, a transmembrane mucin, has been demonstrated a potential prognostic and metastatic marker in breast cancer. However, there have been a limited number of studies addressing the predictive and prognostic features of MUC1 in African breast cancer. This study aims at addressing the expression profiles of MUC1 and other biomarkers in Ghanaian breast cancer and determines its predictive and prognostic characteristics, in relation to other clinicopathological features. Methods. Haematoxylin and eosin (H&E) slides of breast cancer cases were reviewed and 203 suitable cases were selected for tissue microarray (TMA) construction and immunohistochemistry. Anti-ER, PR, HER2, Ki-67, and MUC1 antibodies were used. Results from the immunostaining were analysed using SPSS version 23. Results. About 59% of cases expressed MUC1. Majority of cases in the study showed a lack of expression of all three traditional markers (29% expressed ER, 10.9% PR, and 20.7% HER2). Ki-67 index were 62.1% (low), 16.5% (moderate), and 21.4% (high). MUC1 expressions among the molecular classes were luminal A (60.7%), luminal B (68.8%), HER2 overexpression (87.5%), and triple negative (56.6%). There were significant associations between MUC1 and HER2 overexpression (p=0.01) and triple negative (p<0.01). Conclusion. The high proportion of breast cancer cases expressing MUC1, as well as its association with the two most aggressive molecular classes, indicate a substantial role in the biology of breast cancer in our cohort, and it is an indication of poor prognosis.

PurposeThe aim of this study was to assess the expression of metabolism-related proteins including glucose transporter 1 (Glut-1), carbonic anhydrase IX (CAIX) and monocarboxylate transporter 4 (MCT4) in breast mucinous carcinoma and to evaluate the implications of the results.MethodsImmunohistochemical staining for Glut-1, CAIX, and MCT4 was performed on tissue sections from 59 cases of mucinous carcinoma to evaluate the association between the expression of metabolism-related proteins and clinicopathologic factors. Mucinous carcinoma was subclassified into type A and type B according to histopathological characteristics.ResultsOf the 59 patients, 35 patients (59.3%) were type A mucinous carcinoma and 24 patients (40.7%) were type B mucinous carcinoma. Stromal expression of MCT4 was significantly associated with a high histologic grade (p=0.022) and type B mucinous carcinoma (p=0.016). There were significant positive correlations between the expression of Glut-1, CAIX and tumoral expression of MCT4 (p<0.05).ConclusionWe assessed the expression of metabolism-related proteins including Glut-1, CAIX, and MCT4 in breast mucinous carcinoma and found that the stromal expression of MCT4 was higher in type B mucinous carcinoma than in type A, which reflected a difference in the tumor microenvironment.

Background: Breast carcinoma is a malignancy that occurs from breast cells that grow and continue to proliferate uncontrollably. Mucin 1 (MUC1) has become a topic of interest in cancer treatment due to its upregulation, which affects the invasion, proliferation, and survival of tumor cells, by reducing cells adhesion and extracellular cell-matrix adhesion. Objective: To analyze the correlation between the immunohistochemical expression of Mucin 1 (MUC1) with histopathological grading and molecular subtypes in invasive breast carcinoma. Methods: Formalin-fixed tissue paraffin blocks from 42 patients with invasive breast carcinoma were used as research samples and Mucin 1 (MUC1) immunohistochemical staining was assessed. The correlation between Mucin 1 (MUC1) expression and histopathological grading and molecular subtypes was analyzed by statistical tests using the SPSS program. Results: Of the 42 samples studied, for the age group, the most found were in the age group 40-49 years and the age group 50-59 years (each with 35.7%), while the age group 20-29 years was the least (2.4%). The most histopathological subtypes were invasive carcinoma of no special type (85.7%), with grade 2 being the most common (42.8%). The most common molecular subtype was luminal B (40.5%). Mucin 1 (MUC1) was expressed in 73.8% of invasive breast carcinoma specimens. Conclusion: There is a significant unidirectional correlation between the immunohistochemical expression of Mucin 1 (MUC1), either with histopathological grading or with molecular subtypes in invasive breast carcinoma (p-value were 0.027 and 0.010, respectively).
