Expression of human chorionic gonadotropin (hCG)/luteinizing hormone receptors and regulation of the cyclooxygenase-1 gene by exogenous hCG in human fetal membranes.

Abstract: The present study characterized hCG/LH receptors from messenger ribonucleic acid (mRNA) to protein and whether exogenous hCG can bind and regulate the expression of the cyclooxygenase-1 (COX-1) gene in human fetal membranes from term pregnancy. Northern blotting showed that fetal membranes contain 6.0, 4.4, 2.4, and 1.4 kilobases of hCG/LH receptor mRNA transcripts. In situ hybridization revealed that amnion, chorion, and decidua contain receptor transcripts. Western immunoblotting and immunocytochemistry show… Show more

“…As it is expressed constitutively in many tissues [Langenbach et al., 1995; Simmons et al, 1989], and traditionally not believed to mediate stimulated biological events, Cox-1 has been considered a ''housekeeping'' gene [De Witt and Smith, 1988;Langenbach et al, 1995;Simmons et al, 1989]. Recently, however, the isoform has proven, in some circumstances, to be regulated [Samet et al, 1995;Smith et al, 1993Smith et al, , 1996Toth et al, 1996]. In endothelial cells, for example, cox-1 levels mirror the state of differentiation [Samet et al, 1995].…”

“…The amniotic epithelium is not only one of the main sources of prostaglandins, especially prostaglandin E 2 (4), but also expresses prostaglandin-biosynthesis enzymes such as phospholipase, prostaglandin synthase, and cyclooxygenase (5). Moreover, these enzymes are regulated by human chorionic gonadotropin (hCG), the receptors of which are found on the amniotic epithelium (6).…”

“…9). COX-1 is constitutively expressed in most cell types and has been considered a housekeeping gene (10), although more recent evidence shows regulated expression in both the inflammatory and reproductive systems (11,12). COX-2 is a dynamic isoform induced by specific stimuli in specific cell types (13).…”
