Expression of Hsp70 in the mud crab, Scylla paramamosain in response to bacterial, osmotic, and thermal stress

Abstract: Hsp70 is involved in immune responses against infectious pathogens, thermal, and osmotic stress. To understand the immune defense mechanisms of the mud crab Scylla paramamosain, genomic DNA, transcript level and antimicrobial activities of Hsp70 were analyzed. Genomic DNA sequence analysis revealed one intron in this gene. Furthermore, six SNPs were detected by direct sequencing from 30 samples in this study. Hsp70 mRNA was expressed in almost all tissues examined. By using the quantitative real-time PCR, the … Show more

“…Proteins such as sodium/potassium ATPases and carbonic anhydrases are known to be regulated by salinity transfers for many species (Havird et al 2013). Our result showing that heat induces a stronger HSP induction than changes in salinity is in line with observations in the mud crab Scylla paramamosain where a HSP70 gene was upregulated up to 2.3 times in hyperosmolarity but up to 25 times upon heat shock (Yang et al 2013).…”

“…Although the promoters of HSP70s and their response to environmental stress have been well studied in vertebrates [35], [36], the corresponding information in invertebrates especially in mollusc is still limited [29], [37]. By now, the promoter elements of HSP70s were only predicted in a few seawater species.…”

“…The expression of inducible HSP70 gene is mainly regulated at transcriptional level depending on the interaction between transcription factor and their putative cis-acting elements in the promoter region of HSP70 genes [22] , [34] . “…The overexpression of HSP70 has also been re-X.N. Bao et al ported in the hemocytes of Scylla paramamosain after high salinity stress (Yang et al, 2013). The expression of HSP90 was induced when Crassostrea hongkongensis (Fu et al, 2011) and Crassostrea gigas (Jo et al, 2008) were subject to salinity stress.…”
