Expression of Heat Shock Protein 70 and Heat Shock Cognate 70 Messenger RNAs in Rat Cortex and Cerebellum After Heat Shock or Amphetamine Treatment

Abstract: The expression of strictly inducible hsp70 mRNAs and constitutively expressed hsc70 mRNAs was compared in cerebellum and cerebral cortex of control rats, heat-shocked rats, and rats made hyperthermic with amphetamine. An hsc70-specific oligonucleotide probe identified a 2.55-kb mRNA in cerebellum and cerebral cortex of all rats. An hsp70-specific oligonucleotide probe identified a 3.05-kb mRNA and a 3.53-kb mRNA in cerebellum and cerebral cortex of heat-shocked and amphetamine-treated rats, but not in control … Show more

“…A similar case was also observed in flesh fly Sarcophaga crassipalpis, where Hsp70 was significantly up-regulated under heat shock in nondiapausing flies, while the cognate Hsc70 was not affected (Rinehart et al 2000). However, there are also reports to show that the transcription of Hsc70 in the rat cortex and cerebellum was up-regulated after heat shock, but the extent of variation is much lower than that of Hsp70 (Miller et al 1991). The rapid reaction of FcHsp70 might protect shrimp from acute stresses, during which a large number of denatured proteins are necessary to be chaperoned with Hsps in the cells, and then to reduce the overload of unfolded proteins in the cells .…”

“…However, HSP expression can be induced in these cells in vitro (40,41) as well as in vivo (42), and HSP expression can be protective in several neuropathologies (30). We have therefore tested the possibility that the HS response can reduce and/or prevent astroglial NOS-2 expression.…”

“…The slides were hy bridized at 37°C for 18 h with 1 x 106 cpm of a 35S-labeled synthetic oligonucleotide (30-mer) probe that corre sponds to highly conserved amino acids 122-129 near the 5'-end of the human hsp70 coding sequence (Hunt and Morimoto, 1985). This sequence has been reported to be specific for the inducible hsp70 gene (Miller et al, 1991) and has been used to show that hsp70 mRNA is not ex pressed at detectable levels in most normal rodent cells (Marini et al, 1990;Nowak and Bond, 1990). After hy bridization, the sections were rinsed in 1 x standard sa line citrate (SSC) (150 mM sodium chloride, 15 mM so dium citrate, pH 7.4) at 52°C for 60 min with several changes of 1 x SSC, dehydrated, and exposed to Kodak SB-5 film for a week.…”
