Expression of CD44s mRNA in gastric carcinoma

Abstract

Objective:To study the expression of CD44s mRNA in the occurrence, development and invasion of gastric carcinoma (GC). Methods: The expressions of CD44s mRNA in 66 cases of GC, 25 cases of superficial gastritis and 25 cases of atypical hyperplasia were examined by in situ hybridization (ISH). Results: There was no expression of CD44s mRNA in the group of superficial gastritis; the positive rate was 20%(5/25) in the group of atypical hyperplasia and 62.12%(41/66) in the group of gastric carcinoma. The positive rate in poor differentiation group was significantly higher than that in well differentiation group (P<0.05), and the positive rate of lymph node metastasis group was significantly higher than that in negative lymph node metastasis group(P<0.05). Conclusion: The expression of CD44s mRNA was related to cell differentiation degree and lymph node metastasis, the activation of CD44s gene was related to strong invasion of cancer cells and poor prognosis.Gastric carcinoma (GC) is one of the mo9st common malignant tumors. CG is in the first place of death among all malignant tumors worldwide [1,2] . In recent years, researches on the molecular mechanism of oncogenesis and metastasis and the correlated markers of GC are important objects in clinical research in oncology [2][3][4] . In order to investigate the relationship between the over expression of CD44s mRNA and the oncogenesis invasion, metastasis, and pathological grade of GC, 66 specimens of GC and 50 specimens of non gastric tissue were examined by in situ hybridization technology. MATERIALS AND METHODS Clinical DataThe specimens of GC gastrectomy from 66 patients (49 males and 17 females) with an average age of 57.91 ⎯⎯⎯⎯⎯⎯⎯⎯⎯⎯⎯⎯⎯⎯⎯⎯ years, median age of 59 years (range from 37-79 years)were collected from the department of tumor surgery, College of the First Clinical Medicine, Hebei North University. Specimens from 25 cases of superficial gastritis and 25 cases of gastritis with atypical hyperplasia were also collected among them there were 38 males and 12 females with an average age of 50 years and median age of 48.5 years(range from 39-65 years). Specimens ProcessingGastrectomy specimens from surgery were fixed immediately for 4 hours with 4% formaldehyde in 0.1 mol/L PBS (pH 7.2 containing 0.1% DEPC), then dehydrated, embedded in paraffin wax, sliced up and stained with haematoxylin and eosin. Cases were diagnosed by two seasoned professor. TNM criteria of WHO was applied to the histopathological classification, lymph node metastasis was observed. The multi tissue sections (7 μm) for in situ hybridization were besmeared to the slices with ploy-lysine and kept at 37℃ for use.
