Expression of adiponectin receptors 1 (AdipoR1) and 2 (AdipoR2) in the porcine pituitary during the oestrous cycle

Abstract: BackgroundAdiponectin, protein secreted mainly by white adipose tissue, is an important factor linking the regulation of metabolic homeostasis and reproductive processes. The biological activity of the hormone is mediated via two distinct receptors, termed adiponectin receptor 1(AdipoR1) and adiponectin receptor 2 (AdipoR2). The present study analyzed mRNA and protein expression of AdipoR1 and AdipoR2 in the anterior (AP) and posterior (NP) pituitary of cyclic pigs.MethodsThe total of 20 animals was assigned t… Show more

“…Lack of correlation between the protein and mRNA expression and enzyme activity may result from transcriptional, post-transcriptional or translation regulation, protein stability or enzyme activity as well as functioning feedbacks, i.e. high protein concentration may suppress mRNA expression or enzyme activity, and a high level of gene expression may diminish the post-transcriptional processes (Kiezun et al 2013. The death receptor-mediated pathway of apoptosis is activated when certain death receptor ligands of the tumour necrosis factor (TNF) family (such as FAS (1996) showed the ability of Fas for inducing granulosa cell apoptosis.…”

“…In addition, T-cadherin has been found to act as a receptor for hexameric and HMW forms of adiponectin (Hug et al 2004, Hui et al 2012. Specifically, AdipoR1 in pig tissues has been reported to be highly expressed (Kiezun et al 2013), which is the reason why we focused on this subtype of AdipoRs. In addition, only Adipor1 knockdown has been found to be able to prevent the acute effects of the globular form of adiponectin on rat cardiac metabolism and oxidation (Palanivel et al 2007).…”

“…Forward and reverse primers for GAPDH were used according to the paper of Nitkiewicz et al (2010). The conditions of the reaction were the same as in our previous study (Kiezun et al, 2013). The relative expression levels of AdipoR1 and AdipoR2 were calculated based on the comparative cycle threshold method (ΔΔCt) (Livak and Schmittgen, 2001) and normalised using the geometrical mean of the expression level of reference genes GAPDH and cyclophilin A.…”
