Expression and Activity of Acid-Sensing Ion Channels in the Mouse Anterior Pituitary

Abstract: Acid sensing ion channels (ASICs) are proton-gated cation channels that are expressed in the nervous system and play an important role in fear learning and memory. The function of ASICs in the pituitary, an endocrine gland that contributes to emotions, is unknown. We sought to investigate which ASIC subunits were present in the pituitary and found mRNA expression for all ASIC isoforms, including ASIC1a, ASIC1b, ASIC2a, ASIC2b, ASIC3 and ASIC4. We also observed acid-evoked ASIC-like currents in isolated anterio… Show more

“…B and C). The expression of ASIC4 in the pituitary gland but not the TG or DRG agreed with previous reports from human (Grunder et al ., ), rat (Akopian et al ., ) and mouse (Du et al ., ) samples. In the CNS, X‐gal‐positive signals were detected in many regions, including the olfactory bulb, cortex, hippocampus, amygdala, thalamus, hypothalamus, cerebellum and spinal cord but not adult DRG (data not shown).…”

“…ASIC1a is known to play an important role for sensing acidosis during ischemic stroke, seizure, fear, panic, and MS, as well as synaptic transmission 7 36 37 38 39 40 . A previous study also showed that blockade of ASIC1a activity would result in activation of the endogenous enkephalin pathway in the CNS and thus produce analgesia for neuropathic pain 41 .…”

“…Using probes that differentiated between ASIC1a and ASIC1b , it has been shown that whereas ASIC1a is expressed in rat DRG, spinal cord and brain, ASIC1b expression is restricted to the DRG [28]. However, reverse-transcriptase PCR (RT-PCR) analysis showed that ASIC1b , along with all other ASIC transcripts, is expressed in mouse anterior pituitary tissue [29] and rat trigeminal mesencephalic nucleus neurons [30], suggesting that ASIC1b is present in certain brain regions. A variety of techniques have been used to demonstrate that both ASIC2a and ASIC2b show diffuse expression throughout the brain in a number of species, as well as the spinal cord and DRG [26, 3133].…”
