Exposure to atrazine alters behaviour and disrupts the dopaminergic system in Drosophila melanogaster

Abstract: Atrazine is an extensively used herbicide, and has become a common environmental contaminant. Effects on dopaminergic neurotransmission in mammals following exposure to atrazine have been previously demonstrated. Here, the effects of atrazine regarding behavioural and dopaminergic neurotransmission parameters were assessed in the fruit fly D. melanogaster, exposed during embryonic and larval development. Embryos (newly fertilized eggs) were exposed to two atrazine concentrations (10μM and 100μM) in the diet un… Show more

“…It’s well known that ATR exposure causes dopaminergic alteration, but less is known about behavioral alteration dopaminergic-related [ 39 , 40 , 41 , 42 , 43 , 44 , 45 ]. Catalepsy test was used to investigate the effect of ATR inhalation on both young and aged mice ( Figure 3 A).…”

“…In flies, dopamine modulates a range of behaviours such as locomotion, learning, sleep and courtship 53 . As found in mammals, flies synthesize www.nature.com/scientificreports/ DA from the aromatic amino acid, tyrosine, via the coordinated action of tyrosine hydroxylase and DOPA decarboxylase 54 . Our data revealed the downregulation of ple in PD flies compared with the control flies.…”

“…The activity of tyrosine hydroxylase was measured spectrophotometrically by the formation of dopachrome, essentially as described earlier (Vermeer et al, 2013;Figueira et al, 2017). On the day of eclosion, flies were snap-frozen in liquid nitrogen, vortexed for 20 s at 4 °C to separate fly heads from bodies, then passed through a household sieve to collect the heads, which were stored at -80 °C unless processed immediately.…”
