Nerve endings on adrenaline cells in male rat adrenal medullas were investigated by quantitative electron microscopy, with sampling at 8 times during a standard light:dark (12:12) photoperiod. Number (N) per unit area of 2 vesicle types (small clear vesicle, SCV; large granular vesicle, LGV) and % of LGV per total vesicle N were determined. Normal (non-operated) animals showed a circadian rhythm (p < 0.005) in mean SCVN and one with nearly opposite phase relations in %LGVN. Doubly sham-operated animals had a similar rhythm in %LGVN but diminished circadian change in SCVN. Pinealectomized animals had a circadian pattern in SCVN, but this differed from that of normals in having a 2nd peak, a delay or phase shift of the primary peak, and an increased amplitude of the rhythmic changes.

“…This theory is also consistent with one of the proposed sites of action of melatonin, i. e., the midbrain reticular formation, as suggested by Anton-Tay et al (1968) and Fraschini et al (1968). Quay (1965) also feels that pineal substances generally act on various neural sites and not merely on specific gonadotropin regulating center of the hypothalamus. The fact that anterior and complete cuts were effective in obviating the influence of darkness on reproduction whereas posterior cuts were much less so, may indicate that the axons of the inhibitory neurons may diffusely enter the MBH with the primary concentration of fibers entering from the antero-lateral aspect.…”

SummaryThe influence of total or partial isolations of the medial basal hypothalamus (MBH) on pineal-mediated gonadal responses was studied in adult male hamsters. Light deprivation, due to exposure of hamsters to light: clark cycles of 1:28 (in hours) or blinding, led to involution of the testes, seminal vesieles and coagulating glands and to a statistically insignificant ,depression in bioassayable pituitary follicle stimulating hormone. The effects of darkness and the pineal on the pituitary-gonadal axis were eompletely prevented by total or anterior hypothalamie disconnections; posterior d~sconneetions negated the responses in less 'than one-half the animals. The following tentative explanations for the results were discussed: 1. the hypothalamie cuts interrupted pathways which pass through the hypothalamus and ,transmit photie information between the eyes and the pineal gland, thus rendering the pir~eal unresponsive to photoperiodie stimuli; 2. the surgical manipulations within the hypothalamus caused a "central sympa'thectomy" which ~nterfered with sympathetic control of pineal biosynthetic activity; 8. the hypo,thalamie cuts ~nterrupted axons of extra-MBH pineal responsive neurons tl~at ,are inhibitory to gonadotropin-regulating cells

Summary. Effects of sham-pinealectomy and pinealectomy on preganglionic nerve endings on adrenomedullary adrenaline cells were investigated electron microscopically. Adult male golden hamsters from the normal, shampinealectomy and pinealectomy groups maintained under 24 h light-dark cycle and constant temperature were used at 28 days after surgery. From conventional electron microscopic specimens, montage photographs made of the adrenaline cell region at a magnification of 11,000 were used for qualitative and quantitative electron microscopic analyses in 14 animals in each experimental group. The preganglionic nerve endings were localized mainly in the following three sites: the basal lamina part, the follicular lumen-junctional intercellular part, and the adrenaline cell-invaginated part. In the latter two parts, nerve endings and fi bers had no envelope frequently, and in the former two parts, nerve endings sometimes showed the invagination complex. The frequency of nerve endings was highest in the follicular lumen-intercellular part, next highest in the basal lamina part and lowest in the A cellinvaginated part.
