Expedition into Exosome Biology: A Perspective of Progress from Discovery to Therapeutic Development

Abstract: Exosomes are membrane-enclosed distinct cellular entities of endocytic origin that shuttle proteins and RNA molecules intercellularly for communication purposes. Their surface is embossed by a huge variety of proteins, some of which are used as diagnostic markers. Exosomes are being explored for potential drug delivery, although their therapeutic utilities are impeded by gaps in knowledge regarding their formation and function under physiological condition and by lack of methods capable of shedding light on in… Show more

“…Many exosomes have these characteristics. In addition, they are small, have extremely low toxicity, and can cause a low rate of long-term accumulation in organs and tissues [66,127,167]. They can also cross various biological fluids and pass through plasma membranes, thus delivering therapeutic compounds into the cytoplasm of target cells [147].…”

“…The exosomal indigenous cargo is mostly rich in proteins, lipids, sugars, and nucleic acids [messenger RNAs (mRNAs), microRNAs (miRNAs), and mitochondrial DNA (mtDNA), etc.] (Jan et al, 2021; Figure 1). Exosomes' functions encompass an elaborative list depending on the origin of cell/tissue.…”

“…In studying these oligonucleotide therapeutics, researchers simultaneously work to overcome challenges that occur in treatments such as non-specific delivery of therapeutic RNAs, stimulation of immunogenicity by delivered RNAs or its carriers, risk of degradation by extracellular nucleases, or finding a suitable vector to deliver the therapeutic agent [69,70,73]. One possible solution to that problem seems to be employing exosomes as carriers of therapeutic factors [64,[74][75][76][77]. Exosomes were reported to effectively deliver therapeutic cargo for cancer treatment [70,77], neurodegenerative diseases [65,78] and virus infections including COVID-19 [79,80].…”
