Exosome mimetics derived from bone marrow mesenchymal stem cells deliver doxorubicin to osteosarcoma in vitro and in vivo

Abstract: Osteosarcoma is a bone tumor with a high incidence in children and adolescents. Chemotherapy for osteosarcoma is limited, and effective targeted drugs are urgently needed to treat osteosarcoma. Exosomes as a natural nano drug delivery platform have been widely studied and proven to have good drug delivery performance. However, the low production of exosomes hinders its development as a carrier. Exosome mimetics (EMs) as an alternative product of exosomes solve the problem of low production of exosomes and main… Show more

“…Exosomes secreted from macrophages are endowed with intrinsic tropism towards inflammatory and tumorous tissues [ 67 , 71 74 ]. Mesenchymal stem cell (MSC)-derived exosomes can be derived from adipose tissue [ 75 77 ], bone marrow [ 78 81 ], umbilical cord [ 82 , 83 ], and human placenta [ 84 ], and are most commonly used for tissue regeneration and wound-healing applications [ 76 , 77 , 85 ]. They also exhibit important immunomodulatory properties [ 86 ].…”

“…( B ) Exogenous loading involves the loading of various cargoes, such as drugs, DNAs, ncRNAs, and proteins, into native exosomes. Methods for loading these cargoes include electroporation, sonication, co-incubation, and extrusion [ 136 , 137 , 138 , 139 , 140 , 141 ]. Endogenous loading involves the packaging of biomolecules via the modification of donor cells [ 133 , 134 , 142 , 143 , 144 , 145 ].…”

“…Umbilical cord blood-derived M1 macrophage exosomes that have been loaded with cisplatin via electroporation show more potent effects against cisplatin-sensitive and -resistant ovarian cancer cells than free cisplatin [ 137 ]. Bone marrow mesenchymal stem cell (BMSC)-derived exosomes that have been loaded with doxorubicin show more potent effects against osteosarcoma than free doxorubicin [ 138 ]. These exosomal mimetics are prepared by the extrusion of BMSCs and show fewer side effects than free doxorubicin [ 138 ].…”

For example, Jiang et al discovered seven hub genes, including ANXA1, FKBP11, SP7, and TPM1, which were more upregulated in MSCs than in highgrade OS cells and FDPS, IFITM5, and SQLE, which were more downregulated.…”
