Excavation of Pid3 Orthologs with Differential Resistance Spectra to Magnaporthe oryzae in Rice Resource

Abstract: Twenty-six orthologs of the rice blast resistance gene Pid3 from cultivated varieties and wild rice accessions distributed in different areas were cloned by allele mining. Sequence analysis showed that while each of the orthologous genes from indica varieties and most wild accessions encodes a complete NBS-LRR protein, each of the proteins encoded by those from japonica varieties and few wild rice accessions presents a premature termination. Eleven of the 26 orthologs were selected for blast resistance testing… Show more

“…Such an approach of allelemining has been deployed in wheat for managing powdery mildew wherein different alleles of the resistance gene Pm3 have been cloned from different accessions of wheat and have been shown to confer resistance (Bhullar et al , 2010). Functional orthologues of the gene Pid3 that confer resistance to rice blast with different resistance spectra have been isolated and cloned (Shang et al , 2009; Lv et al , 2013; Xu et al , 2014).…”

“…The wheat powdery mildew resistance gene Pm3 with 17 identified functional alleles is a remarkable example of natural variations present in GenBank accessions and can be efficiently utilized for conferring broad-spectrum disease resistance. For rice blast resistance gene, functional orthologous have been found in wild rice O. rufipogon accessions (Lv et al, 2013; Xu et al, 2014; Ashkani et al, 2015) defining their utility in widening the genetic base of cultivated rice varieties. A novel allele of PSTOL1 gene is identified in O. glaberrima (CG14) and being transferred to the NERICAs (New Rice for Africa) cultivars using allele-specific markers.…”

“…Lv et al, (2013) isolated an ortholog of Pid3 from a common wild rice accession A4 (O. rufipogon) using sequencingbased allele mining, called Pid3-A4, with confirmation of different amount of resistance spectra. Thereafter, Xu et al (2014) isolated 26 orthologs of Pid3 from cultivated and wild rice accessions and evaluate their respective blast resistance by gene transformation and blast inoculation. Another study performed to help formulate a good allelemining set has a heuristic program with SSR genotype information from 4046 rice accessions, and the allelemining the set established is being tested and verified (Zhao et al, 2010).…”
