Evolutionary relationships of members of the genera Taphrina, Protomyces, Schizosaccharomyces, and related taxa within the archiascomycetes: Integrated analysis of genotypic and phenotypic characters

Abstract: To study the phylogeny and evolution of archiascomycetes, we determined the full sequence of the nuclear 18S rRNA gene from 14 Taphrina species and 2 Protomyces species, and the partial sequence of Schizosaccharomycesjaponicus var.japonicus. The sequences were phylogenetically analyzed by the neighbor-joining, maximum parsimony, and maximum-likelihood methods. We also looked at their principal phenotypic characters and genotypic character.

“…Sjamsuridzal et al (1997) assumed that this strain might have been misidentified as Taphrina based on morphological, chemotaxonomic or physiological and biochemical characteristics, in addition to the sequence analysis. Since 'Taphrina' maculans CBS 427.69 is not available for distribution from the CBS, this strain has not been compared with CBS 973 T in our laboratory, but it is assumed that CBS 973 T , 'Taphrina' maculans CBS 427.69 and 'Taphrina' californica CBS 374.39 (Sjamsuridzal et al, 1997) are closely related strains.…”

“…Gjaerum, 1964;Bacigálová, 1997). More recently, molecular methods (namely sequencing of the 18S rRNA gene) have been used to unveil phylogenetic relationships among Taphrina species and other members of the archiascomycetes, but this study involved only a limited number of species represented by single strains (Sjamsuridzal et al, 1997). Mix (1949Mix ( , 1954 and Gjaerum (1964).…”

“…However, there has been some confusion in the literature dealing with Taphrina due to the inadvertent study of strains of ascomycetous or basidiomycetous yeasts misidentified as Taphrina species (e.g. Heath et al, 1982;Sjamsuridzal et al, 1997;Moore, 1998).…”

“…The primers used in this study were the same as described in Sjamsuridzal et al (1997). PCR products were cloned by the Invitrogen ® TA cloning kit (Andres, 1993).…”
