SUMMARYWe have generated a polyclonal antibody (CKG2) against native chicken gizzard ecto-ATPase for immunolocalization and immunoprecipitation. Active ecto-ATPase is immunoprecipitated from solubilized chicken and rat membranes and shown to be localized to the plasma membrane of the chicken smooth muscle cells. This antibody is specific for the ecto-ATPases, since the more abundant chicken stomach ecto-apyrase is not recognized in immunoprecipitation, western blot or immunolocalization analyses. The CKG2 antibody cross-reacts with mammalian (rat) ectoATPase in western blots, with testis being the most abundant source. Interestingly, when the same rat membranes are analyzed by western blot under non-reducing conditions, the 66 kDa ecto-ATPase is not recognized, instead a 200 kDa protein is detected, previously postulated to be an oligomer of ecto-ATPase. However, this 200 kDa cross-reacting protein is not related to the ecto-ATPases, but is instead an immunoglobulin binding protein, comprised of 50 kDa subunits.

The isolated cells were obtained from hog thyroid glands treated with dispase. More than 95 % of the cells obtained were intact and viable immediately after preparation, and the cell viability did not change during incubation in the experimental conditions. ATP added to the external medium of whole cell suspensions was hydrolyzed in the presence of various divalent cations, especially Mg, and the rate of hydrolysis of ATP was not significantly different between the Mg-ion system and the completed ion system (Mg+ Na +K). When whole cell suspensions were disrupted with homogenizer, the hydrolysis of ATP was markedley increased by adding Na plus K. But there was no difference in the Mg-ion system between cell homogenates and whole cell suspensions.ADP, AMP and adenosine as reaction products were found in the reaction mixture which resulted from the hydrolysis of ATP by whole cell suspensions.Our data suggest that Mg-ATPase in the thyroidal isolated cells is an ectoenzyme whose active site(s) are exposed to the external surface of plasma membrane, and that ATP is finally hydrolyzed to adenosine via ADP and AMP by the enzyme(s).

Objectives: Exposure to chromium, the transition element found in many compounds of earth's crust, leads to various health hazards including cancer, dermatitis, damage of liver and kidneys, and reproductive toxicity in both male and female. Toxic effect of chronic exposure to chromium at low environmentally relevant dose is recently recognized and less studied. In the present study, we evaluated the effect of subchronic exposure to low doses of chromium VI (K 2 Cr 2 O 7 ) on various hematological and biochemical parameters in male albino rats. Methods:Male albino rats (160±10 g) received 1/25, 1/50, and 1/100 of reported oral LD 50 dose of chromium VI (K 2 Cr 2 O 7 ) for 90 consecutive days. After treatment, the blood was collected for analysis of various hematological and biochemical parameters.Results: Exposure to chromium VI at 1/25 LD 50 dose caused decrease in body weight gain and reduction in the weight of the reproductive organs along with increase in the weight of the liver and kidney. Numbers of red blood cells, white blood cells, platelets, and hemoglobin concentration reduced with increase in clotting time. The total sperm count and motility was also reduced in exposed rats. There was a decrease in plasma protein and albumin concentration, with increase in plasma glucose and cholesterol level. The activity of enzyme alanine aminotransferase and enzyme aspartate aminotransferase was stimulated in treated rats. Conclusion:The results of the present study revealed that chronic low-dose exposure to chromium VI resulted in a toxic effect in experimental male albino rats.
