Evidence for the occurrence of an ecto-(adenosine triphosphatase) in rat epididymal spermatozoa

Abstract: Intact spermatozoa from rat cauda epididymis possess a Mg2+-dependent ATPase activity that hydrolyses externally added [gamma-32P]ATP. The ATPase reaction was linear with time for approx. 6 min and there was no detectable uptake of ATP by these cells. The ATPase activity of the whole spermatozoa was not due to leakage of the intracellular enzymic activity, contamination of the broken cells or any possible cell damage during incubation and isolation of spermatozoa. The activity of the enzyme was strongly inhibi… Show more

“…rat spermatozoa was described almost twenty years ago [26,27] as having some of the classic properties of the E-type ATPases. However, some of the spermatozoa ecto-ATPase activity described earlier is sensitive to vanadate inhibition, unlike the prototypical 66 kDa ecto-ATPase.…”

“…Similar criteria have been met for phosphodiesterase (Smoke et al, 1981), 5'-nucleotidase (Stefanovic et al, 1976), protein kinase (Chiang et al, 1979 ;Majumder, 1981), ATPase (Trams and Lauter, 1974 ;DePierre and Karnovsky, 1974), p-nitrophenyl phosphatase (DePierre and Karnovsky, 1974) and leucine aminopeptidase (Nagaoka and Yamashita, 1981). In these ecto-enzymes, ecto-ATPase has previously been reported in several cell types (Majumder and Biswas, 1979 ;Harlan et al, 1977 ;Smolen and Weissmann, 1978 ;Bosmann et al, 1968: Chambers et al, 1967Modolell and Moore, 1967). However, the function of ecto-ATPase has not been clarified.…”
