Evidence for Rapid Oxidative Phosphorylation and Lactate Fermentation in Motile Human Sperm by Hyperpolarized 13C Magnetic Resonance Spectroscopy

Abstract: Poor sperm motility is a common cause of male infertility for which there are no empirical therapies. Sperm motility is powered by adenosine triphosphate but the relative importance of lactate fermentation and Oxidative Phosphorylation (OxPhos) is debated. To study the relationship between energy metabolism and sperm motility we used dissolution Dynamic Nuclear Polarization (dDNP) for the first time to show the rapid conversion of 13C1-pyruvate to lactate and bicarbonate, indicating active glycolytic and OxPho… Show more

“…Lactate was previously reported as the main metabolite produced by human spermatozoa [39]. In addition, human spermatozoa are able to further metabolize lactate into pyruvate and acetate [39,40], which is corroborated by our results that show spermatozoa acetate production after glucose treatment. We were also able to identify and quantify malate in the media.…”

“…Organic acids are produced by the breakdown of amino acids and fatty acids, and the degradation of such metabolites generates energy substrates for tricarboxylic acid (TCA) cycle and respiratory chain [38]. The presence of organic acid suggests that bull spermatozoa have active energy metabolism [8, 39, 40]. In addition, organic acids play crucial roles during anabolism by providing C-atom backbones [38].…”

“…The concept of spermatogenic lactate shuttles was introduced by L. Bruce Gladden in his classic review paper (Gladden, 2004). Gladden's review unified extant data in the literature and anticipated subsequent results in the field of fertility so important for human reproduction, veterinary medicine, and animal husbandry agriculture (Boussouar and Benahmed, 2004;Reynolds et al, 2017). Gladden's treatment of the subject has stood the test of time even as the technologies, including magnetic resonance spectroscopy (MRS), have become more sophisticated (Reynolds et al, 2017).…”
