Ruminant energy supplementation with vegetable oils or fats has been standing out worldwide and oil palm processing has been receiving growing interest. This study assessed the effect of supplementation with saturated and unsaturated fatty acids from the palm oil industry on the lipid profile of seminal plasma and of the sperm membrane, as well as on the morphological and functional characteristics of raw and cryopreserved buffalo semen. Twelve purebred Murrah bulls ( Bubalus bubalis ) were assigned to the experimental groups and fed diets for 120 days with no added lipids (CONT, four bulls), or with an extra amount of 3% lipids from crude palm oil (PALM, four bulls), or from palm oil deodorizer distillate (PODD, four bulls). Semen was collected and cryopreserved every 15 days. The lipid composition of membranes and semen quality were determined after collections. Lipid supplementation did not impact feed intake (P>0.05). Diet enrichment with PALM increased the linoleic acid (C18:2,ω6) in seminal plasma. Lipid supplementation did not increase the polyunsaturated fatty acids in the sperm membrane composition, but significantly increased the lignoceric acid (C24:0). Cryopreserved semen of the supplemented bulls presented higher progressive motility (60.2 vs. 67.9 vs. 65.2%; P<0.05) and sperm viability detected by eosin-nigrosin staining (61.1 vs. 69.4 vs. 67.8%; P<0.05). Palm oil reduced major sperm defects in both raw (12.2 vs. 9.3 vs. 13.2%; P<0.0001) and cryopreserved semen (12.4 vs. 9.4 vs. 11.2%; P<0.0001). The lipids added to the diet did not impact the population of spermatozoa with intact plasma and acrosomal membranes (PI-/PSA-), but significantly increased the percentage of spermatozoa with high mitochondrial potential (25.6 vs. 31.5 vs. 32.0%; P=0.008). The results suggest that lipid supplementation based on crude palm oil or palm oil deodorizer distillate can be safely used to feed buffalo bulls and may increase sperm attributes related to male fertility.

The effects of adding mixed chicken and Japanese quail egg yolks (EYs) to the cryodiluent on the quality of ram semen before freezing and post-thawing were evaluated. Additionally, the composition of chicken and quail egg EYs and their mixture were analyzed for results explanation. The semen was collected from rams (n = 5) and extended with cryodiluent containing the EY of chicken, quail or their mixture (1:1). The extended semen was chilled slowly to 5 °C within 2 h and equilibrated for 2 h, before frozen on the liquid nitrogen vapor and cryopreserved at −196 °C. The straws were evaluated before freezing and post-thawing for sperm motility, vitality and abnormality besides plasma-membrane and DNA integrities. The moisture, ash, protein, and fatty acid (FA) contents of chicken EY, quail EY and their mixture were analyzed. Sperm vitality, plasma membrane integrity and DNA integrity before freezing were significantly (P < 0.05) higher in quail EY than chicken EY and mixed EYs cryodiluent. The chicken EY extender significantly improved the vitality, plasma membrane and DNA integrities of post-thawed ram semen in comparison with quail EY or mixed EYs extenders. While, the post-thawing sperm abnormalities was lower (P ≤ 0.05) in quail EY than chicken EY and mixed EYs cryodiluent. The post-thawing sperm motion kinetics parameters were higher in quail EY than chicken EY and mixed EYs cryodiluent. The highest percentages of moisture, ash, saturated fatty acids (SFAs) and monounsaturated fatty acids (MUFAs) were detected in quail EY had. While, the highest percentages of fat, protein and polyunsaturated fatty acids (PUFAs) were detected in chicken EY. In conclusion, using of chicken EY can improve total motility, vitality, plasma membrane integrity and DNA integrity of cryopreserved ram semen. While, using of quail EY can improve sperm abnormalities and kinetic motion parameters of cryopreserved ram semen. Mixing chicken and quail EYs added no value for post-thawing ram semen parameters.

Aim: To evaluate the bacterial composition of collared peccary semen and foreskin mucosa, and to verify the sensitivity of isolates to antimicrobials used in semen conservation and to Aloe vera gel, which is an alternative external cryoprotectant. Methods and Results: Nine foreskin mucosa and ejaculate samples from adult animals were used. Sperm characteristics and bacterial load were evaluated in fresh semen. The preputial mucosa and semen bacterial isolates were identified and tested against five concentrations of each antimicrobial (streptomycinpenicillin and gentamicin) and A. vera gel. Corynebacterium sp. and Staphylococcus sp. were isolated in greater numbers than others in both semen (64Á10 and 20Á51%, respectively) and the foreskin mucosa (60Á60 and 24Á25%, respectively), and ranged from 0Á4 to 21 9 10 5 colony-forming units (CFU) per ml. The average load of Corynebacterium sp. was negatively correlated (P < 0Á05) with the sperm membrane integrity (r = À0Á73055) and curvilinear velocity (r = À0Á69048). Streptomycin-penicillin and gentamicin inhibited most microorganisms , and A. vera showed lower antimicrobial activity. Conclusion: Several Gram-positive bacteria are present in semen and foreskin mucosa of collared peccary, and the benefits of using primarily penicillinstreptomycin and gentamicin antimicrobials in the bacterial control of diluted semen of these animals are strongly indicated. Significance and Impact of the Study: This study provides insight into the reproductive microbiota of captive male-collared peccary. This work provides a theoretical basis to assist reproductive biotechnologies for ex situ conservation of the species.
