Aim: Alveolar bone resorption, often occurring after tooth extraction, can be minimized through socket preservation. This process uses a combination of Moringa leaf extract and demineralized freeze-dried bovine bone xenograft (DFDBBX) that is expected to generate both transforming growth factor-beta 1 (TGF-β1) expressions as a transcription factor associated with osteoblast differentiation and osteocalcin accelerating alveolar bone formation. This research aimed to analyze the role of the combination of Moringa leaf extract and DFDBBX induced in socket preservation when generating TGF-β1 and osteocalcin expressions. Materials and Methods: The left mandibular incisors of 56 Cavia cobaya were extracted and divided into four groups subjected to different socket preservation treatments. The first group treated with polyethylene glycol, the second group with DFDBBX, the third group with Moringa leaf extract, and the fourth group with a combination of DFDBBX and Moringa leaf extract. The C. cobaya were examined on days 7 and 30, after which the specimens were sacrificed and examined using an immunohistochemical technique. The resulting data were then analyzed using one-way ANOVA and Tukey's honestly significant difference tests. Results: There was a significant difference in TGF-β1 and osteocalcin expressions between the groups ( P < 0.05). The highest mean amount of TGF-β1 and osteocalcin was found in the fourth group on both days 7 and 30. Conclusions: The combination of Moringa leaf extract and DFDBBX can effectively generate TGF-β1 and osteocalcin expressions during the preservation of tooth extraction sockets.

BackgroundImmunohistochemistry (IHC) is a well-established, widely accepted method in both clinical and experimental parts of medical science. It allows receiving valuable information about any process in any tissue, and especially in bone. Each year the amount of data, received by IHC, grows in geometric progression. But the lack of standardization, especially on the post-analytical stage (interpreting and reporting of results), makes the comparison of the results of different studies impossible.MethodsComprehensive PubMED literature search with a combination of search words “immunohistochemistry” and “scoring system” was performed and 773 articles describing IHC results were identified. After further manual analysis 120 articles were selected for detailed evaluation of used approaches.ResultsSix major approaches to the interpretation and presentation of IHC analysis results were identified, analyzed and described.ConclusionsThe overview of the existing approaches in evaluation and interpretation of IHC data, which are provided in the article, can be used in bone tissue research and for either better understanding of existing scoring systems or developing a new one. Background: Preservation of sockets is a procedure aimed to reduce bone resorption after tooth extraction. One of the most commonly used xenograft materials is demineralized freeze-dried bone bovine xenograft (DFDBBX). Meanwhile, one of the key regulations in osteoclast genesis process is RANKL bond. A decrease in the number of RANKL expressions can suppress the osteoclast genesis process so that bone resorption can be prevented. The combination of Moringa leaf extract and DFDBBX, as a result, is expected to decrease the number of RANKL. Purpose: This study aimed to measure RANKL expressions in tooth extraction socket treated with Moringa leaf extract combined with DFDBBX. Methods: Fifty six Cavia cobaya rats were divided into eight groups. The first group was a control group with PEG administration onto their extraction sockets. The second group was a treatment group with DFDBBX administration. The third group was a treatment group with Moringa leaf extract administration. The fourth group was a treatment group induced with a combination of DFDBBX and Moringa leaf extract. Examination then was performed on days 7 and 30. After 7 and 30 days, those Cavia cobaya rats were executed and tested with immunohistochemical techniques. Further research data collected then were tested with one-way ANOVA. Results: There were significant differences between the control group and the groups induced with the combination of Moringa leaf extract and DFDBBX. On days 7 and 30, the groups induced with the combination of Moringa leaf extract and DFDBBX had the lowest number of RANKL expressions. Conclusion: The combination of Moringa leaf extract and DFDBBX can decrease the number of RANKL expressions in Cavia cobaya rats on the day 7 and day 30 after tooth extraction.
