Evaluation of the automated hematology analyzer Sysmex XT-2000iV™ compared to the ADVIA® 2120 for its use in dogs, cats, and horses

Abstract: The automated laser-based hematology analyzer Sysmex XT-2000iV™ providing a complete blood cell count (CBC) and 5-part differential has been introduced in large veterinary laboratories. The aim of the current study was to determine precision, linearity, and accuracy of the Sysmex analyzer. Reference method for the accuracy study was the laser-based hematology analyzer ADVIA® 2120. For evaluation of accuracy, consecutive fresh blood samples from healthy and diseased cats (n = 216), dogs (n = 314), and horses (n… Show more

“…Although the CVs for RBC and platelet‐related variables measured by LaserCyte were < 10%, they are moderate compared with the variations obtained in previous reports using other cytometry laser‐based instruments in horses and small animals . The higher CV for RBC count observed with the LaserCyte could be due to mild fluctuations in the noise level of the laser technology, as previously reported .…”

“…This new and sophisticated equipment provides a more complete, rapid, and accurate CBC than either manual methods or impedance systems. Additionally, some of these instruments can provide supplementary data, such as reticulocyte counts, population scatter plots, analyses based on a species‐specific software, granularity index, CD4‐/CD8‐positive lymphocyte counts, estimations of stem‐cell numbers by quantifying surrogate cells, and reticulated platelet counts . Briefly, in impedance systems, cell counts and sizing are based on the detection and measurement of changes in electrical resistance produced by each cell as it passes through a small aperture with an electrode located on each side.…”

“…Meta-analysis and assessment of TE obs based on data obtained from hematologic method validation studies A meta-analysis of the hematology analyzer validation data for canine and feline blood published between 2005 and 2013 was performed [13][14][15][16][17][18] , and the observed total error (TE obs ) for variables determined with commonly used hematology analyzers were calculated. Inclusion criteria for acceptable data were availability of CV from the replication experiments, and the regression analysis equation.…”

“…Overall, 4 hematologic method validation studies were included in the meta-analysis evaluating the performance of the Sysmex XT 2000iV [13][14][15][16] , CellDyn 3500 17,18 and ADVIA 2120. 13,14 Recorded CV from replication experiments, slope and intercept from regression analyses, calculated bias, and calculated TE obs are listed in Table 3 and 4, respectively. For several variables, we determined TE obs > TE a .…”

“…It is beyond the scope of this article to assess these machines, but a list of references is given that may be helpful for determining their utility. [7][8][9][10][11][12] Reference intervals should be established for your practice population, as these will vary in different areas of the country and also with the breeds that are present within a practice area. If you are using a reference laboratory, then it is important to understand how their reference intervals were established and whether they fit with your practice.…”
