Evaluation of the association between a single-nucleotide polymorphism of bone morphogenetic proteins 5 gene and risk of knee osteoarthritis

Abstract: Background:Osteoarthritis (OA) is a chronic degenerative disorder probably affected by both genetic and environmental causes. Bone morphogenetic proteins (BMPs) are bone-derived factors that can induce new bone formation. Single-nucleotide polymorphisms (SNPs) of BMP5 gene alters the transcriptional activity of the BMP5 promoter that has been involved in OA susceptibility. This case–control study investigated the association of rs1470527 and rs9382564 SNP of BMP5 gene with susceptibility to knee OA (KOA).Mater… Show more

“…Sharma et al . [ 10 ] analyzed genotypic frequencies in Indian men and women. They found a 5.41- and 16.42-fold increased risk of KOA in men and women, respectively, when carrying CT genotype, while the risk is 3.42 and 5.08 fold, respectively, when carrying TT genotype compared with CC genotype.…”

“…The mRNA expression of aggrecan (AGC) and Col2A signifying chondrogenic differentiation was highest where sustained BMP2 is delivered via BMP-coacervate [50]. Both BMP -2 and 7 appeared promising disease-modifying agents for OA patients and are currently under phase-II trial [51,52]. Human MSC, including ADMSCs, treated with BMP-4, 6, and 9 modulate in vitro cartilage development [46].…”

“…Human MSC, including ADMSCs, treated with BMP-4, 6, and 9 modulate in vitro cartilage development [46]. Recently, in a case-control study, a significant association of rs1470527 and rs9382564 polymorphism of BMP -5 gene with human OA-Knee has been demonstrated; this also may be a potential therapeutic target for OA [52].…”

“…BMP5 is a bone morphogenetic protein known to promote chondrocyte differentiation, and mutations within this gene have been associated with osteoarthritis. 75 The upregulation of BMP5 appears to be necessary in the physiological repair pathway required for RCT healing. One high-quality study found BMP5 to be upregulated in torn supraspinatus or infraspinatus tendon tissue that successfully healed after surgical repair of RCTs at 1 year, as compared with tendons with failed healing.…”
