Evaluation of the Antioxidant Activity of Copper(II) Complexes containing Tris‐(hydroxymethyl)aminomethane (TRIS) Units

Abstract: The structural analysis and the SOD-like activity of Cu II complexes [(Cu)(C 16 H 18 N 3 O 5 )]ClO 4 (1) and [(Cu) 4 (C 36 H 52 N 8 O 12 )] (2) are described with ligands obtained from the condensation of picolinaldehyde and 2-formylpyrrole with tris(hydroxymethyl)aminomethane

“…The evaluation of catalytic activity mimicking that of SOD was conducted using the photoreduction method of nitroblue tetrazolium (NBT) in a buffer solution (PBS; pH = 7.8 . This analysis shows the inhibition of the reaction between the superoxide ion and NBT and therefore the non‐production of formazan chromophore compound identified spectrophotometrically and generated reaction of superoxide radicals with NBT indicator.…”

“…Schiff bases like H 3 L1, derived from the reactions among TRIS and pyridyl or quinolinealdehydes, linked to polyoxometallates or lanthanides with a SMM response for the Dy III case, have been recently characterized. However, 3d complexes, containing cycled oxazolidines like H 3 L3 instead of the corresponding Schiff base, appear to be the most common reaction, and some mononuclear Cu II or dinuclear Fe III compounds derived from the pyridyl ligands and several mononuclear or low nuclearity 3d systems derived from the quinoline precursors have also been reported. Formation of the oxazolidine ring requires the nucleophilic attack of one of the alkoxo arms on the iminic C atom and the transfer of the proton to the N atom, Scheme .…”

“…Reaction of pyridine- or quinolinealdehyde with tris(hydroxymethyl)aminomethane (TRIS) is one of the reactions that, usually in the presence of metallic cations, tends to produce oxazolidines instead of the corresponding Schiff bases. The number of coordination complexes derived from 2-pyridinecarboxaldehyde or quinolinealdehydes, often mononuclear systems, is limited but has been characterized for several 3d cations over the last several years. However, this catalyzed formation of the oxazolidine has not been reported yet when using salicylaldehyde or the related o -vanillin as the starting carbonyl source.…”

“…The superoxide dismutase (SOD)-like activity was measured according to Canabarro et al, (2016) using SOD Assay Kit (Sigma-Aldrich ® , Darmstadt, Germany). Briefly, extracts (20 µL) were mixed with WST (Water Soluble Tetrazolium) and enzyme working solutions (prepared by diluting 15 μL of enzyme solution with 2.50 mL of dilution buffer; 200 μL), and then incubated in the dark at 37 °C for 20 min.…”
