Evaluation of platelet mitochondria integrity after treatment with Mirasol pathogen reduction technology

Abstract: There is no evidence from these studies that Mirasol PRT treatment alters PLT mitochondrial structural and functional integrity immediately after treatment and during PLT storage. An increased demand for ATP may be the driving force for observed increases in both the glycolytic flux and the oxidative metabolism observed in treated PLTs.

“…Clinical studies have demonstrated that pH value is a simple parameter which indirectly indicates the condition of platelets and correlates with in vivo platelet recovery. 19,20 It has been determined that the mean platelet volume decreases in all components stored at room temperature, which confirms the satisfactory condition of cells.…”

“…Many authors claim that parameters such as aggregation capacity or HSR represent the in vivo hemostatic functions of platelets, but the most reliable, conclusive method of demonstrating PC transfusion effectiveness would be to evaluate the patient's clinical state following the transfusion of PRT-treated PCs. 20,30 Independently of the above-described in vitro studies, each regional blood transfusion center involved in PC inactivation also performs routine quality control of inactivated PCs and monitors the components. If the percentage of components within a normal range falls below 75%, corrective measures are launched which consist of monitoring all stages of inactivated PC preparation.…”

“…Marschner/Goodrich spite the fact that mitochondrial DNA is fragmented by the treatment [11], the wavelength of UV light used in the process is distinct from the one where cytochrome C, FMN and FAD absorb (370-450 nm) [46], resulting in intact electron transfer processes within mitochondrial membranes. Low oxygen levels in the treated products are a simple measurement and indication of functional mitochondria, since oxygen is consumed and ATP produced during mitochondrial respiration [44,45]. Additional evidence that mitochondria are intact after Mirasol treatment comes from the use of PAS, in which acetate is used as a substitute for glucose.…”
