Evaluation of Neonatal Streptozotocin Induced Diabetic Rat Model for the Development of Cataract

Abstract: Type 2 diabetes (T2D) generally follows prediabetes (PD) conditions such as impaired fasting glucose (IFG) and/or impaired glucose tolerance (IGT). Although studies reported an association of IGT or IFG with cataract, the experimental basis for PD associated cataract is not known. Hence, we evaluated neonatal streptozotocin (nSTZ) induced rat model to study PD associated cataractogenesis by injecting STZ to two-day old rats. While majority (70%) of nSTZ injected pups developed IGT (nSTZ-PD) by two months but n… Show more

“…injection of STZ on the second day of birth at dose level 90 mg kg −1 , developed insulin resistance. 26 In the present study, the extracts showed statistically significant reduction in the blood glucose levels at p < 0.05 on day 30 ( Table 2 ). A statistically significant decrease in the glycated hemoglobin levels was observed at p < 0.05.…”

“…From the viewpoint of the hepatic L-alanine production, thus, the oral intake of L-glutamine can be regarded as an additional health benefit in the case of type 2 diabetes, which also includes increased glucose uptake and diminished insulin resistance [ 5 , 9 ]. The main characteristics of type 2 diabetes in humans were also detected in the model (neonatal streptozotocin induced diabetic rat) used in the present study, as for example mild fasting hyperglycemy, high insulin levels and some degree of insulin resistance [ 55 , 56 , 57 ]. However, the behavior of livers from type 2 diabetic rats with respect to L-glutamine metabolism was essentially that one observed with livers from healthy rats if one excepts the already mentioned increased L-alanine production.…”

“…Other studies have similarly shown that different parameters of oxidative stress and antioxidant defense are impaired in DR. Among these parameters, retinal SOD activity and iPF 2 α levels have been shown to decrease and increase, respectively, in animal models of DR …”
