Evaluation of Matrix-Assisted Laser Desorption Ionization-Time-of-Flight Mass Spectrometry in Comparison to 16S rRNA Gene Sequencing for Species Identification of Nonfermenting Bacteria

Abstract: Nonfermenting bacteria are ubiquitous environmental opportunists that cause infections in humans, especially compromised patients. Due to their limited biochemical reactivity and different morphotypes, misidentification by classical phenotypic means occurs frequently. Therefore, we evaluated the use of matrix-assisted laser desorption ionization-time-of-flight mass spectrometry (MALDI-TOF MS) for species identification. By using 248 nonfermenting culture collection strains composed of 37 genera most relevant t… Show more

“…Cells were extracted using a standard formic acid/acetonitrile method (Mellmann et al 2008). Extracts (1.2 ll) were spotted onto a sample target in duplicate, left to airdry, and were overlaid with 1.2 ll of matrix, containing alpha-cyano-4-hydroxycinnamic acid (HCCA) as a saturated solution in 50 % acetonitrile and 2.5 % trifluoracetic acid.…”

“…As the database grows, the concordance with the gold standard identifications increases steadily. At this time, it is at 98 % for bacteria and yeast species [40][41][42][43][44]. Even the difficulties are now predictable; foreseeable, reliable difficulties can be circumvented.…”
