Objective Acanthamoeba is a free-living and opportunistic amoeba that the potential of this parasite to convert to a cyst, making its treatment difficult. In this study, we investigated the effect of microwave radiation on Acanthamoeba cysts in vitro. Methods Acanthamoeba cysts were reproduced in the culture medium. We divided 16 test tubes containing cysts into two groups. The contents of 8 tubes were irradiated with continuous microwave in the time range of 0-120 s, and the next 8 tubes were irradiated with repetitive microwave in the range of 0-360 s. The mortality of cysts at the end of irradiation was recorded and compared with control. Results Continuous and repetitive irradiation of microwave resulted in mortality of 100 (%) of Acanthamoeba cysts during 120 and 360 seconds, respectively. In continuous mode, parasite mortality in all groups was significantly different from the control group (P<0.0001). But in the repetition pattern only in groups that had been irradiated for more than 60 seconds, parasite mortality was significantly different from the control group (P<0.0001). Conclusion The change of radiation mode from continuous to repetitive, reduces the thermal effects of the microwaves but does not affect the mortality rate of Acanthamoeba cysts. Therefore, probably the impact of microwaves was not only caused by the increase in ambient heat but also its another property is involved in the death of Acanthamoeba.

AIM: Aprevious study confirmed the effect of microwaves on protoscolices in vitro. In the current study, the effect of shortwave diathermy (SWD) on the protoscolices of hydatid cysts was investigated. MATERIALS AN DMETHODS: The hydatid cyst was collected from the slaughterhouse and its contents were drained sterilized. SWD therapy was done in continuous mode. The experiment was conducted based on electrode area, power of device, and exposure time.GraphPad and SPSS software was used for data analysis. RESULTS: The study results showed that the two main factors of time exposure and power of the machine could significantly effective in the mortality rate of protoscolices compared to control (p < 0.0001). Scolicidal activity of SWD in 450 w was 4.5%, 8.3%, and 10%, respectively, after 2, 5, and 10 min with ΔT = 0.5°C, 14°C, and 16°C. The results of the study showed that the simultaneous effect of two factors of power and time exposure increased significantly death rate of protoscolices compared to control (p < 0.05). CONCLUSION: Since the application of SWD has limited risks, ease of use, and relative low cost and it can induce hyperthermia in depth of tissues, continuous SWD can be used as a tool for preoperative in surgery. However, it is likely that scolicidal activity can be observed by changing the device mode in pulse short wave at a lower temperature and also the possible side effects need further investigation in vivo.

The present study investigated the effect of the Neodymium-Doped Yttrium Aluminum Garnet (Nd: Y3AL5G12) Laser eradiation 1000 mw, against infection with secondary hydatid disease in BALB/c mice by inoculating the animals with protoscoleces of Echinococcus granulosus exposed to laser irradiation, for various periods 40,60,120 and 240 minutes, with viability of 70%,57%,50% and 40%, respectively, contrasted to the control set (mice inoculated with unexposed protoscoleces) along three months, depending on the evaluation of the total count of WBCs and assessment of acquired cell-mediated immunity, demonstrated by delayed type hypersensitivity test (DTH). The results showed considerable increase (p<0.01) in the total WBCs counting, in mice injected with irradiated protoscoleces, up to 13962 cell/cm 3 , in comparison to the control group 5420 cell/cm 3 , after 120 minutes, two months post infection. Moreover, there was considerable excess (p<0.01) in the foot pad thickness in treated mice, 2.94 mm after 120 minute (24hour post antigen injection), two months post infection, 2.86mm after 120 minutes (24 hours post antigen injection), 1 month post infection and 2.64 mm, 120 minutes (24 hours post antigen injection), three months post infection, compared with the control group 1.18mm, 1.80mm and 1.72mm, respectively.Results revealed that irradiation of Echinococcus granulosus protoscoleces with laser had a significant influence on the production of WBCs and stimulation of delayed-type hypersensitivity reaction in mice.
