Characterization studies provide essential information for the conservation and use of germplasm in plant breeding programs. In this study, 103 Capsicum frutescens L. accessions from the Active Germplasm Bank of Embrapa Hortaliças, representative of all five Brazilian geographic regions, were characterized based on morphological characteristics and microsatellite (or simple sequence repeat - SSR) molecular markers. Morphological characterization was carried out using 57 descriptors, and molecular characterization was based on 239 alleles from 24 microsatellite loci. From the estimates of genetic distances among accessions, based on molecular characterization, a cluster analysis was carried out, and a dendrogram was established. Correlations between morphological and molecular variables were also estimated. Twelve morphological descriptors were monomorphic for the set of C. frutescens accessions, and those with the highest degree of polymorphism were stem length (14.0 to 62.0 cm), stem diameter (1.0 to 4.2 cm), days to flowering (90 to 129), days to fruiting (100 to 140), fruit weight (0.1 to 1.4 g), fruit length (0.6 to 4.6 cm), and fruit wall thickness (0.25 to 1.5 mm). The polymorphism information content for the SSR loci varied from 0.36 (EPMS 417) to 0.75 (CA49), with an overall mean of 0.57. The correlation value between morphological and molecular characterization data was 0.6604, which was statistically significant. Fourteen accessions were described as belonging to the morphological type tabasco, 85 were described as malagueta, and four were malaguetinha, a morphological type confirmed in this study. The typical morphological pattern of malagueta was described. Six similarity groups were established for C. frutescens based on the dendrogram and are discussed individually. The genetic variability analyzed in the study highlights the importance of characterizing genetic resources available for the development of new C. frutescens cultivars with the potential for various niche markets.

Old traditional Serbian pepper landraces (Nizača, Lokošnička and Turšijara), grown in South Serbia and produced by seed sowing or transplanting, were evaluated in this study. Besides the basic morphological properties and composition, the features characterizing fruit color and pericarp anatomy were determined in order to analyze and systematize the traits relevant from the aspect of traditional end use of examined landraces. Landraces Lokošnička and Nizača, characterized with higher force needed to puncture fruit skin, larger number of mesocarp layers, thicker endocarp and thicker collenchyma tissue, followed with higher total soluble solids content are more suitable for intended traditional use than Turšijara. Production from transplants should be favored in order to obtain more uniform, darker red fruit color, ASTA value, higher TSS content as well as higher yields. There are genetic differences of the some quality differences between the cultivars. Lokošnička has higher mesocarp layers, thicker endocarp and collenchyma tissue. Practical Applications Pepper fruits dried in traditional way slowly in a draught are used in preparation of local ethno‐gastronomic specialities, with special position in local customs related to dietary habits during the spring Orthodox fasting period. Intact fruit skin is also the prerequisite for successful utilization in traditional culinary specialties‐stuffed dry pepper fruits previously soaked in water that remain whole during cooking and baking. Structure and texture of pepper fruit skin and pericarp can consequently be considered as one of the key aspects of technological quality of pepper based on which landraces were selected for described traditional preservation technique. The research for answers to this technological challenge was the motivation of this study, which evaluated the impact of different predrying process (type of landraces, production methods) on qualities of pepper fruit. The data shown would provide scientific rationales for obtaining high‐quality whole fruit pepper as material for stuffing.

Genetic Diversity Analysis of 41 Chili Pepper Genotypes [Capsicum annuum L.] Based on SSR Markers

<p>Chili (Capsicum annuum L.) is an important horticultural crop which possesses high genetic diversity. Their genetic diversity needs to be assessed through molecular marker-based analysis to facilitate chili breeding scheme. The objective of the study was to analyze the genetic diversity of 41 chili genotypes using Simple Sequence Repeat (SSR) markers. Eleven SSR markers were used to amplify all genotypes through PCR technique, visualized on Polyacrylamide Gel Electrophoresis (PAGE). The data were analyzed using NTSYSpc version 2.1 and PowerMarker version 3.25 softwares. Results showed that the average of<br />SSR alleles was 7.9 with a range of 4-11 alleles per marker. All SSR markers showed PIC value >0.5, indicating their suitable use for chili genetic studies. Cluster analysis of 41 genotypes generated three main groups according to coefficient similarity of 77.4%. The genetic clusters appeared as reflection of the genetic background of each genotype. In addition, the SSR markers used can identify potential parent as indicated by the genetic distance between genotypes. The highest genetic diversity (0.698) was demonstrated by combinations of Yuni × Ayesha, Yuni × Ayesha 2, and Yuni × Namira. The estimated genetic diversity produced by SSR markers in this study should be useful as guidance for chili breeders to select suitable parents for new cultivar development for high productivity and resistance to diseases.</p>
