This study evaluated the stability of resin-dentin interfaces treated with glutaraldehyde-containing agents, and assessed collagen degradation in dentin matrices treated with Gluma. Microtensile bond strength (μTBS) was evaluated 24 h and 6 months after treatment with three desensitizers (Gluma Desensitizer, Gluma Desensitizer Power Gel, and MicroPrime G) and two etch-and-rinse adhesives (Comfort Bond & Desensitizer and iBond TE). Demineralized beams of human dentin were treated with water or Gluma, and the degradation of collagen in these beams was assessed by quantification of the dry mass loss and of the amount of hydroxyproline released from hydrolyzed specimens after 1 or 4 wk. All groups demonstrated significant reduction in μTBS after 6 months, except for Gluma Desensitizer and iBond TE groups, which showed decreases of 7.2% and 10.8%, respectively. The most prevalent failure mode was 'mixed'. Significantly less hydroxyproline was released from Gluma-treated beams than from control beams after 4 wk. Beams treated with Gluma yielded significantly less dry mass loss than did beams in the control group. Collagen cross-linking with glutaraldehyde-containing agents may assist in the stabilization of resin-dentin bonds by reducing the amount of collagen solubilized from dental matrices in the hybrid layer. In turn, this may contribute to the preservation of adhesive interfaces.

Background: This study aimed to evaluate possible cytotoxic effects to gingival epithelial cells exposed to children toothpastes containing different detergent. Methods: Tissues required fort he isolation of human gingival epithelial cells were obtained by biopsy during the extraction of the impacted third molar tooth. Toothpaste solutions of different concentrations were prepared from five different children’s toothpastes with different detergent contents. Isolated gingival epithelial cells were stimulated with experimental groups consisting of toothpaste solutions (Colgate, Sensodyne, Splat, Nenedent, Perlodent) at different concentrations and a control group consissting of complete Dulbocco’s modified eagle medium. After the experiments, cell viability was evaluated using flow cytometry. Data analysis were done using One Way ANOVA test and Tukey post-hoc test. Results: In all experimental groups, there was a decrease in live cell rates and an increase in dead cell rates due to increased concentration. The statistically highest live cell ratios were detected in Splat’s toothpaste solutions after the control group and the group with the lowest viability values was determined in Colgate group (p<0.05). Conclusions: According to the results of the study, it was observed that toothpastes containing SLS affected the viability of cells more negatively than toothpastes with other detergent contents.

Bioactive glass plus laser phototherapy as promise candidates for dentine hypersensitivity treatment

Treatments for dentine hypersensitivity (DH) may produce positive effects, though do not have lasting results. We investigated the reparative potential of stem cells derived from deciduous teeth (SHEDs) in response to components delivered from substances used in the treatment of the DH, associated or not to laser phototherapy (LPT), to stimulate dentine formation. SHEDs were submitted to substances delivered from a laboratorial P-rich bioactive glass [57SiO -26CaO-17P O (wt %)] or a commercially available desensitizer (Gluma® Desensitizer), associated (or not) to LPT (InGAlP diode laser, 660 nm, 0.028 cm , 20 mW, 5 J/cm , 7 s, contact mode). Biomaterial characterization was performed by X-ray diffraction, scanning electron microscopy and the particle size was evaluated by dynamic light scattering. SHEDs proliferation and differentiation were analyzed by MTT and Alizarin Red staining, respectively. The conditioned media used in these tests were evaluated regarding their pH and the ionic concentration changes due to ions leached from the bioactive glass (BG). BG majority presented a non-crystalline solid structure and mixed particle sizes characterized by the agglomeration of nanoparticles. Cultures treated with BG alone or in association to LPT showed improved cell growth in relation to Gluma (p < 0.05). Gluma was cytotoxic in all tested conditions, regardless irradiated or not. BG associated to LPT induced intense mineral matrix formation. In conclusion, BG releases ionic dissolution products able to promote SHEDs differentiation. BG associated to LPT improves SHEDs proliferation and differentiation in vitro, and may be a promise therapeutic approach for the DH treatment. © 2015 Wiley Periodicals, Inc.
