Antibacterial, cytotoxicity and metabolite profiling of crude methanolic extract from andaliman (Zanthoxylum acanthopodium) fruit. Biodiversitas 21: 4147-4154. The local community in North Sumatra has utilized andaliman fruit (Zanthoxylum acanthopodium) as spices for traditional cuisines because it has a unique flavor. Information on the antimicrobial activity of Z. acanthopodium fruit against aquaculture pathogens and its bioactivity against leukemia cell lines are limited. The purposes of this study were to evaluate the antimicrobial activity of Z. acanthopodium fruit against Tenacibaculum maritimum, Vibrio alginolyticus, V. anguillarum, V. harveyi that are known as pathogens in aquaculture; to determine cytotoxic property against murine P388 leukemia cells; and to characterize its metabolites profile. The sample was extracted using methanol by the maceration method. Antibacterial assay was conducted by Kirby-Bauer disc diffusion method; while cytotoxicity assay using the XTT method. Proximate analysis showed that Z. acanthopodium fruit contained 63.41% of moisture, 24.73% of crude fiber, 9.81% of crude protein, 6.90% of ash, and 2.55% of crude fat. Several phytochemical components were detected, such as alkaloid, flavonoid, tannin, triterpenoid, and steroid. The GC/MS analysis indicated the presence of various compounds from terpenoid and terpenes derivatives. This study indicated that Z. acanthopodium fruit was not potential as antibacterial agents against the aquaculture pathogens; however, the methanol extract showed cytotoxic potential with IC50 19.14 µg/mL against murine P388 leukemia cells.

Zanthoxylum acanthopodium DC. is a widely used traditional medicinal plant to treat fever, flu, stomachache, traumatic injury, and mosquito bite in tropical and subtropical Asia. This study aimed to investigate the antifungal, anti-inflammatory, antiviral, and larvicidal activities of its fruit essential oil. The essential oil sample from China (EOZC) was mainly composed of limonene (29.78%) and β-myrcene (26.65%), while the sample from Myanmar (EOZM) was dominated by Terpinen-4-ol (43.35%). Both essential oils showed antifungal activity, with 90% minimum inhibitory concentration (MIC90) values ranging from 26.3 to 499 μg/mL. By obviously inhibiting nitric oxide (NO) in RAW 264.7 cells, EOZC (IC50, 16 μg/mL) showed comparable anti-inflammatory activity to the positive control L-NMMA (IC50, 12.2 μg/mL). EOZM showed significant antiviral activity against the dengue virus with an IC50 value of 13 μg/mL. Additionally, both EOZC and EOZM demonstrated dose-dependent larvicidal activity against Aedes albopictus, with LC50 and LC90 values ranging from 45.8 to 144.0 μg/mL. Our results contribute a theoretical foundation for the further application of Zanthoxylum acanthopodium DC. as an antifungal and anti-inflammatory ingredient in the pharmaceutical industry and further indicate that it has the potential to be developed as a new source of natural and eco-friendly medicine for the prevention and treatment of dengue virus.

Background: Hepatocellular carcinoma (HCC) or hepatoma is the most common primary liver cancer and causes many deaths. Andaliman extract functions as an antioxidant against several cancer cells such as breast cancer cells and cervical cancer cells. Aim: To investigate the effect of andaliman fruit extract (Zanthoxylum acanthopodium) on the anticancer activity of HepG2 liver cancer cell lines. Methods: This research is an experimental study, exploratory laboratory to prove andaliman fruit extract to inhibit proliferation rate, inhibit growth, and triggers apoptosis of HepG2 liver cancer cell lines in vitro. HepG2 liver cancer cell lines were cultured on a suitable and controlled medium to grow. The n-hexane extract, ethyl acetate extract and ethanol extract of andaliman fruit were dissolved in dimethylsulfoxide (DMSO) solution, vortexed and made up with MK-RPMI. Further dilution was carried out to obtain a test solution with a concentration of 500 ug/mL, 250 ug/mL, 125 ug/mL, 62.5 ug/mL and 31.2 ug/mL. The cytotoxicity test results of andaliman fruit against HepG2 cells was carried out using the MTT assay. Results: In this study we found that the IC50 of andaliman fruit extract was 122,656 µg/mL. Conclusions: The results show that there is a cytotoxic activity from the andaliman extract on HepG2 liver cancer cell. Our further study is to assess and compare the anticancer activity from the andaliman extract and doxorubicin on HepG2 liver cancer cell.
