Evaluation of basal micronucleus frequency and hexavalent chromium effects in fish erythrocytes

Abstract: Hexavalent chromium (Cr [VI]) genotoxicity was studied using fish micronucleus analysis in peripheral blood erythrocytes from Pimephales promelas, the fathead minnow. Forty-five- to 60-d-old fish were used to assess the spontaneous level of genotoxic damage. The genotoxic effect of Cr (VI) obtained from potassium dichromate (K2Cr2O7) in tests performed for 7-, 14-, and 21-d exposure periods was estimated. Significant micronucleated erythrocyte (MNE) induction was detected in fish exposed for 7 d to 2.5 mg/L of… Show more

“…The researchers also proposed the mean of 2.0 ± 3.3 x 10 -4 for micronucleus erythrocytes. This frequency baseline is similar to those presented in several fish species studied fish species (Al-Sabti & Metcalfe, 1995;Grisolia & Starling, 2001;Lemos et al, 2001;Bolognesi et al, 2006). Considering this baseline value, A. jacuhiensis from rio Uruguai basin present in both environments and seasons analyzed values higher than the one pointed as baseline for Lemos et al (2008).…”

“…This is in agreement with some (De Lemos et al, 2001;Cavas et al, 2005) studies that have reported decrease in the number of MN in fish erythrocytes after 14-21 of exposure. This could mean that DNA repair occurred after the day 14 onwards.…”

“…Induction of micronuclei was reported previously for fish exposed to Cd [Ayllon and Garcia Vazquez, 2000;Sanchez-Galan et al, 2001;Zhu et al, 2004;Arkhipchuk and Garanko, 2005;Cavas et al, 2005], Cu [Zhu et al, 2004;Arkhipchuk and Garanko, 2005;Cavas et al, 2005], Cr [Al-Sabti et al, 1994;de Lemos et al, 2001;Zhu et al, 2004], and Pb Ferraro et al, 2004]. Therefore, it is plausible that elevated concentrations of heavy metals could have contributed to the elevated MN and NA frequencies produced by the downstream Berdan River water samples.…”
