Background: Acanthus ilicifolius L. is admired for its traditional usage in the folk medicine for the treatment of numerous diseases including allergy and helminthiasis in various parts of the planet. The ethanolic extract of the aerial parts of this shrub (EEAI) was investigated in the present study for its major phytochemical groups, antiallergic activity, anthelmintic activity, cytotoxicity and for acute toxicity. Methods: Antiallergic activity was carried out using Toluene 2, 4-diisocyanate (TDI)-induced allergic mice model by assessing various symptoms of allergic rhinitis like sneezing, scratching, swelling and watery rhinorrhea as well as counting the total and differential leukocytes profile of blood. The paralysis and death time of parasites, Haemonchus contortus (Nematoda) and Paramphistomum cervi (Trematoda) were used for anthelmintic activity test. Mortality of mice was counted to evaluate the acute toxicity whereas the mortality of brine shrimp was taken into account to assess cytotoxic potential of the extract. Results: Phytochemical screening of the extract demonstrated the presence of alkaloids, phenolic compounds, tannins, flavonoids, glycosides, saponins, steroids and triterpenoids. Oral pretreatment of the extract significantly ameliorated the TDI-induced allergic symptoms like sneezing (p < 0.05), scratching (p < 0.05), swelling and watery rhinorrhea in experimental mice. The extract also reduced the differential count of leukocytes in the blood which was increased due to induction of allergic conditions through TDI sensitization. In anthelmintic activity test the extract revealed a dose dependent decrease in the relative index of paralysis and death for both H. contortus and P. cervi parasites and thus indicated the extract to be parasiticidal at higher concentrations. In brine shrimp lethality bioassay of toxicity assessment, the LC 50 of the standard drug vincristine sulfate was 0.43 μg/mL whereas the extract showed the LC 50 as 44.57 μg/mL indicating a promising cytotoxicity of the extract. In acute toxicity study the highest dose 3 g/kg failed to show any mortality in Swiss albino mice and thus confirmed the safety of the extract for in vivo administration. Conclusions: The present study corroborated the traditional uses of the aerial parts of Acanthus ilicifolius L. in allergic diseases and in helminthiasis.

Traditional medicinal plants used for respiratory disorders in Pakistan: a review of the ethno-medicinal and pharmacological evidence

Respiratory disorders are a common cause of malady and demise in Pakistan due to its remoteness, cold and harsh climatic conditions as well as scarce health care facilities. The people rely upon the indigenous plant resources to cure various respiratory disorders. The primary objective of this review was to assemble all available ethno-medicinal data of plants used for respiratory disorders in Pakistan. Pharmacological activity of these plants (based upon published scientific research), distribution, diversity, use, preparation methods, economical value, conservation status and various available herbal products of some plants have also been explored. This study scrutinized various electronic databases for the literature on medicinal plants used in Pakistan to treat respiratory disorders. A total of 384 species belonging to 85 families used to treat respiratory disorders in Pakistan has been documented. Cough was the disorder treated by the highest number of species (214) followed by asthma (150), cold (57) and bronchitis (56). Most of the plants belongs to Asteraceae (32) and Solanaceae family (32) followed by moraceae (17), Poaceae (13), and Amaranthaceae (13) with their habit mostly of herb (219) followed by Shrub (112) and tree (69). Traditional healers in the region mostly prepare ethno medicinal recipes from leaves (24%) and roots (11%) in the form of decoction. Among the reported conservation status of 51 plant species, 5 were endangered, 1 critically endangered, 11 vulnerable, 14 rare, 16 least concern, 3 infrequent and 1 near threatened. We found only 53 plants on which pharmacological studies were conducted and 17 plants being used in herbal products available commercially for respiratory disorders. We showed the diversity and importance of medicinal plants used to treat respiratory disorders in the traditional health care system of Pakistan. As such disorders are still causing several deaths each year, it is of the utmost importance to conduct phytochemical and pharmacological studies on the most promising species. It is also crucial to increase access to traditional medicine, especially in rural areas. Threatened species need special attention for traditional herbal medicine to be exploited sustainably.

BACKGROUND:Ziziphus jujuba belongs to family Rhamnaceae widely distributed in subtropical and tropical countries. It is used traditionally for several pharmacological purposes including anti-inflammation, antidiarrhoeal and antibacterial, as well as tonic and sometimes as hypnotic (sedative).AIM:To determine the in vivo antidiarrhoeal, antibacterial and anti-inflammatory activities of Z. jujuba fruit ethanolic extract.METHOD:The fruit was macerated and extracted by 95% (v/v) ethanol. The antidiarrhoeal activity was evaluated using castor oil and Escherichia coli induced diarrhoea mouse model. The antidiarrhoeal and antibacterial activity was investigated at graded doses (400-1200 mg/kg). The anti-inflammatory effects were tested using the carrageenan-induced paw oedema in female Wistar rats. Rat’s treatment groups received tragacanth, 100 mg/kg diclofenac sodium, 800 mg/kg, 1200 mg/kg or 1600 mg/kg of an ethanolic extract of Z. jujuba (EEZJ). All treatment groups were fed with the compounds one hour before carrageenan injection at of rat’s paw. Also, the EEZJ was further analysed by HPLC-PDA system for identification of the presence of betulinic acid and quercetin.RESULTS:EEZJ different doses did not show inhibitory activity against castor oil induced diarrhoea except for the higher (1200 mg/kg) dose. However, the frequency of defecation of stools and watery stool were reduced significantly when compared to control group (P ≤ 0.05 and P ≤ 0.01 respectively), resulted in overall 67% inhibition of diarrhoea. Our anti-inflammatory results demonstrated that EEZJ was able to inhibit the carrageenan-induced paw oedema in rats to a significant degree (p ≤ 0.05) and the paw volume and thickness of both left and right paw were affected compared to the negative control group.CONCLUSION:EEZJ possesses antidiarrhoeal and antibacterial activity in a dose depending manner and may provide a pharmacological basis for its clinical use in diarrheal diseases. The activity may partially be due to the presence of betulinic acid and quercetin.
