Evaluation of a Novel Automated Protocol for the Collection of Peripheral Blood Stem Cells Mobilized with Chemotherapy or Chemotherapy Plus G-CSF Using the Fresenius AS104 Cell Separator

Abstract: Forty-seven peripheral blood stem cell (PBSC) collections were carried out on patients mobilized with chemotherapy and 63 on patients mobilized with chemotherapy plus G-CSF (Filgrastim), using the Fresenius AS104 cell separator and a novel automated PBSC collection protocol. As expected, cell yields were significantly higher in the series mobilized using chemotherapy plus G-CSF. The low platelet and red blood cell contamination permitted freezing of the apheresis product without further manipulation, other tha… Show more

“…The collection of PBSC can be performed on various cell separator devices operating with semiautomated or automated apheresis programs [9, 10, 11]. The aim of the present study was to evaluate collection yields of conventional-volume COBE Spectra AutoPBSC leukaphereses and to demonstrate clinical significance of collected PBSC following HDCT.…”

“…Other reports described CD34-CE between 48 and 87% at preapheresis peripheral blood CD34+ cell counts of approximately 30/µl, using the cell separators Fresenius AS104, Haemonetics MCS-3P, COBE Spectra (protocol 4.7) and/or Fenwal CS3000 [9, 10, 11, 19, 20, 21]. Some authors reported better CD34-CE at a preapheresis level of circulating CD34+ cells <50/µl [13, 22].…”

“…The study was approved by the Hospital Human Investigation Review Board and written informed consent was obtained from all patients. Patients underwent PBPC collection by leukapheresis using the Fresenius AS104 (Fresenius, St Wendel, Germany) blood cell separator and a previously described automated collection protocol (Pierelli et al, 1993). The collection procedures were started when CD34 cell concentration exceeded 20 cells/l.…”
