Evaluating anti-oxidant potential of ganoderic acid A in STAT 3 pathway in prostate cancer

Abstract: Evaluating anti-oxidant potential of Ganoderic acid A in STAT 3 pathway in Prostate cancer. Molecular docking and ADMET activities of different isoforms of ganoderic acid on STAT 3 pathway were performed by Maestro 9.6 (Schrödinger Inc). The ganoderic acid A is best-docked among isoforms which analyses the expression level of antioxidant and STAT 3 pathway in PC-3 cells. The receptor-based molecular docking reveals the best binding interaction of SH2 domain of STAT3 and ganoderic acid A with GScore (-6.134), k… Show more

“…During our initial screening, GA-A stood out among the 12 purified GA monomers because of its optimal in vitro activity in the MDCK cyst model. GA-A has been found to suppress the proliferation, metastasis and adhesion of breast cancer cells, prostate cancer cells, pancreatic cancer cells, etc., in previous studies [ 35 37 ]. The mechanisms by which GA-A inhibits cell proliferation and viability and accelerates cell apoptosis mainly involve a decrease in Cdk4, which targets the STAT3 and Wnt proteins [ 35 37 ].…”

“…Mn-SOD is a kind of superoxide dismutase located in the mitochondrial matrix which takes manganese ion as the active center, can protect the cells from oxidative stress by converting superoxide anion into less toxic hydrogen peroxide [ 23 ]. We further studied the effect of bibenzyl derivatives in D. officinale on Mn-SOD and SIRT3 activity in vitro.…”

“…Similarly, increased STAT-3 activation and its DNA binding occur in the prostate of mice fed a high-fat diet [57]. In human PC cells, the inhibition of STAT-3 results in the inhibition of cell proliferation and a significant decrease in cell viability [58].…”
