ABSTRACT:The study evaluated qualitative and quantitative characteristics of Amazonian jundiá semen. The experiment was carried out in February 2015 at Cia do Peixe, a company located in Goiás state, Brazil, that specialises in fish fry production. Three male L. marmoratus farmed in dugout ponds, weighing 1.34 ± 0.29 kg and measuring 39.33 ± 1.15 cm in length, were tested. The specimens were captured and transferred to a 1000-L tank at the Reproduction Laboratory of the abovementioned company. Water temperature was measured at 7:00 a.m. and 5:00 p.m., and the fish were fed twice daily. For semen collection, the fish were captured from the tank using a dip net and constrained with a wet cotton towel. Their eyes were covered and their urogenital papilla was cleaned and dried with paper towel. Semen was collected in microcentrifuge tubes that were immersed in ice and analysed immediately. On average, the sperm motility rate was 83.33%, motility duration was 5.33 min, sperm concentration was 1,795,833 cells/ mm 3 , and vigour was 4. The morphological analysis of the sperm revealed tail anomalies in 17.66%, midpiece anomalies in 20%, and head anomalies in 5.33%. Based on qualitative and quantitative characteristics, L. marmoratus semen was shown to be suitable for use in artificial reproduction procedures.

RESUMO -Para determinação da dose inseminante ideal na fertilização artificial de ovócitos de jundiá cinza (Rhamdia quelen), foram selecionadas duas fêmeas com papila urogenital avermelhada, abdômen arredondado e liberação de ovócitos de cor e tamanho uniforme e quatro machos que apresentavam liberação de sêmen sob pressão da papila genital. As fêmeas e os machos receberam duas doses de extrato de hipófise de carpa (doses inicial e final de 0,5 e 0,25 mg.kg -1 e de 5,0 e 2,5 mg.kg -1 , para fêmeas e machos, respectivamente). As duas fêmeas produziram 169 g de ovos contendo 1.141 ovócitos.g de ovócitos -1 e os machos, um total de 23,6 mL de sêmen. Do sêmen dos quatro machos foi realizado um pool e analisadas as seguintes características: concentração espermática (1,97x10 10 espermatozóides.mL -1 ), índice de sobrevivência espermática (96%) e morfologia espermática (66,9% de espermatozóides normais). No ensaio de fertilização, utilizou-se delineamento experimental inteiramente casualisado, composto de cinco tratamentos (6x10 3 , 6x10 4 , 6x10 5 , 6x10 6 , 2x10 7 espermatozóides.ovócito -1 ) e quatro repetições. Após 8 horas de incubação, foram analisadas as taxas de fertilização para cada tratamento. A melhor relação espermatozóide.ovócito -1 foi de 89.497 espermatozóide.ovócito -1 , o que provocou taxa de fertilização máxima teórica de 86,68%. SummaryThe aim of this study was to evaluate if hormonal treatment causes changes in the morphology of matrinxã Brycon amazonicus spermatozoa. Twelve males were randomized into three treatment groups: 1, Ovaprim™ (0.5 ml/kg); 2, carp pituitary extract (CPE; 2.0 mg/kg); and 3, NaCl solution 0.9% - control group (0.5 ml/kg); with four replicates. Morphological sperm analysis was performed using an optical and scanning electron microscope. The percentage of normal spermatozoa (49.6±4.6% to 60.8±2.8%), with primary (26.1±6.4% to 45.3±4.5%) and secondary abnormalities (4.4±2.9% to 13.9±3.5%) did not differ significantly between treatment groups. There were no significant differences between treatments in relation to the primary abnormalities found in the head (10.5±3.8% to 25.5±6.3%), the midpiece (0.1±0.1% to 0.2±0.2%) and in the flagellum (18.9±3.8% to 15.5±3.1%), as well as in the secondary abnormalities, located in the head (0.8±0.4% to 7.8±4.6%) and in the flagellum (2.2±1.1% to 6.1±1.5%). When each abnormality was evaluated individually, only the percentage of degenerated head was higher in the CPE-induced group (24.9±5.9%) than the control group (7.2±3.1%). We concluded that the use of pituitary extract of carp or Ovaprim™ under the conditions of this experiment does not influence the percentage of sperm abnormalities in B. amazonicus.
