Estimation of Interleukin-1β Promoter (−31 C/T and −511 T/C) Polymorphisms and Its Level in Coronary Artery Disease Patients

Abstract: Interleukin-1β (IL-1β) is an inflammation-causing cytokine that exerts several unique biological effects and could lead to future adverse events of CAD. The piece of work presented herein is aimed at investigating possible association of IL-1β levels to its polymorphic site viz. -511 and -31 at promoter region in Saudi CAD patients. The study included 155 confirmed CAD patients and 80 healthy control individuals both men and women. Concentration of IL-1β in the patients' serum was measured by ELISA method. For… Show more

“…In our earlier studies, we reported non‐significant associations of SNPs in promoter region of IL‐6, IL‐18, and IL‐1β genes with Saudi CVD patients . However, the present study reports a significant association between polymorphism in IL‐10 promoters at −1082 and −819 sites with Saudi CVD patients.…”

“…In our earlier studies, we have reported a significant elevation in different inflammatory mediators such as IL‐6, IL‐18, and IL‐1β in Saudi CVD patients . In this study too, we observed a significant elevation in IL‐10 level in CVD patients and other cardiac events (Table ).…”

“…NLRP3 activation and consequent inflammation‐causing cytokine interleukin‐1β (IL‐1β) secretion are well‐characterized in various types of cells. Although its known protective effect against infections and its importance for proper immune response following tissue damage; excessive inflammasome activation has been linked to plentiful diseases, including cancer and neurodegenerative disorders …”

“…In contrast to IL-18 gene, IL-1b gene has TATA box, which contains the core promoter region and two important binding sites for the leucine zipper containing transcription factor (TF), CCAAT/enhancer binding protein-b, and the myeloid-specific Ets domain containing TF PU.1 (5,35). It has been reported that IL-1b gene contains several SNPs, including -511 T/C, -1464 C/G, -2023 C/A, -3737 G/A, -31 T/A, +289 C/T, and +3953 C/T, and may be associated with altered expression of the cytokine (20,50,51).…”
