Establishment of three cell lines from Chinese giant salamander and their sensitivities to the wild-type and recombinant ranavirus

Abstract: Known as lethal pathogens, Ranaviruses have been identified in diseased fish, amphibians (including Chinese giant salamander Andrias davidianus, the world’s largest amphibian) and reptiles, causing organ necrosis and systemic hemorrhage. Here, three Chinese giant salamander cell lines, thymus cell line (GSTC), spleen cell line (GSSC) and kidney cell line (GSKC) were initially established. Their sensitivities to ranaviruses, wild-type Andrias davidianus ranavirus (ADRV) and recombinant Rana grylio virus carryin… Show more

“…ADRV infection caused a serious systemic hemorrhagic disease and led to more than 90% mortality in Chinese giant salamanders [ 6 , 7 , 9 ]. Although significant progresses have been achieved in understanding the pathogenesis and genome structure of ADRV as well as host immune responses against viral infection [ 9 , 10 , 11 , 12 , 13 , 14 , 15 ], no effective treatment for the virus is available.…”

“…1 ). RGV can replicate and cause cytopathic effects in cultured CGS thymus cell (GSTC) line which was the first established cell line of CGS [ 38 , 39 ]. The two viruses with high similarity provided us useful materials to investigate the interspecies infection of ranaviruses.…”

“…Epithelioma papulosum cyprinid cells (EPCs) and Chinese giant salamander thymus cells (GSTCs) [29] were cultured in Medium 199 supplemented with 10% fetal bovine serum (FBS) at 25 °C. They were used for all work needing cells, except the co-immunoprecipitation (co-IP) assay.…”
