Establishment of Human Colon Carcinoma Lines in Nude Mice

Abstract: After subcutaneous inoculation into nude mice of 24 human colon adeno-carcinomas, growth, defined as histopathologically confirmed tumor growth which has been passed, was observed in 13 cases (54%). Tumors from metastatic sites showed higher take rates (58%) than tumors from primary sites or recurrent tumors (50%). Nine continuous tumor lines were established (69% of growing tumors) with metastatic tumors establishing more readily (100% of growing tumors) than primary tumors (40%). The average period in primar… Show more

“…In other human cancer types (i.e., breast cancers), pejorative clinical and biological factors are associated with an increased in vivo tumor take (6,23,24). As reported with other cancers (25), in vivo tumor growth in this study population constituted a predictive factor for overall survival and, in nonmetastatic patients, for metastasis-free survival.…”

“…With the large amount o f material studied, the data have permitted the analysis of growth in primary transplant and passages in relation to tumor category, tumor type (primary site/metastasis/recurrent tumor) and grade of differentiation. Previous papers have reported on colon carcinomas (3) and tumors o f the breast, lung, head and neck, female genital tract, bone and soft tissue (4). The present paper extends the analysis to include other gastro intestinal tumors, germ cell-primitive cell tumors, kidney tumors, lower urinary tract tumors, malignant melanomas and a number o f métastasés from unknown primary sites, from which growth and establishment o f tumor lines have been accomplished.…”

“…Several publications from this laboratory on xenotransplantation of human tumors to the nude mouse have been concerned with the growth and histopathology o f the tumors and, more recently, with the establishment of tumor lines (3)(4)(5)15). With the large amount o f material studied, the data have permitted the analysis of growth in primary transplant and passages in relation to tumor category, tumor type (primary site/metastasis/recurrent tumor) and grade of differentiation.…”

“…In addition, serially transplantable xenografts were frequently established: 20 of 26 fragments grafted in mice led to established xenografts. Human colon carcinoma grafting into immunodeficient mice to obtain serially transplantable xenografts has frequently been reported but rarely used for secondary in vitro cell line establishment (17)(18)(19). The rate of xenograft and/or subsequent cell line success reported here seems to be as high as those reported by other investigators.…”
