Establishment and characterization of two primary breast cancer cell lines from young Indian breast cancer patients: mutation analysis

Abstract: Two novel triple negative breast cancer cell lines, NIPBC-1 and NIPBC-2 were successfully established from primary tumors of two young breast cancer patients aged 39 and 38 years respectively, diagnosed as infiltrating duct carcinoma of breast. Characterization of these cell lines showed luminal origin with expression of epithelial specific antigen and cytokeratin 18 and presence of microfilaments and secretary vesicles, microvilli, tight junctions and desmosomes on ultra-structural analysis. Both the cell lin… Show more

“…A recent study of Pandrangi S L from New Delhi, India established Two novel triple negative breast cancer cell lines, NIPBC-1 and NIPBC-2 from primary tumors of two young breast cancer patients aged 39 and 38 years respectively, diagnosed as infiltrating duct carcinoma of breast. Screening for mutations in BRCA gene revealed presence of three heterozygous polymorphisms in exon 11 of BRCA1 gene in both the cell lines [24]. The allocation of BRCA1 mutations recognized among Indian high-risk patients is similar to what has been observed in other populations.…”

“…Developing thoroughly characterized cell lines, from this rare cancer type is central to accelerating research and development. [1519] We report the establishment and characterization of the first cell line derived from primary untreated salivary duct carcinoma. Importantly, the cell line shared lineage and molecular characteristics and both the full-length AR and the AR-V7 isoform with the parent tumor.…”

“…While cancer cell lines maintain many features of the tumor tissue (Neve et al, 2006;Vargo-Gogola and Rosen, 2007;Van Staveren et al, 2009), there are some difficulties with using them. For instance, the majority of them have been isolated from a metastatic tumor of older patients and many of these cell lines have been in culture for decades (Ku et al, 2013;Pandrangi et al, 2014). In addition, the genetic diversity of tumor cells may cause unpredictable tumor behavior and treatment failure.…”
