Establishment and Characterization of Acute B-Cell Lymphocytic Leukemia Cell Line Showing (8; 14) and (14; 18) Chromosome Translocation

Abstract: Cell line KHM-2B expressing two oncogene products, c-myc and bcl-2, was established from a patient with actute lymphocytic leukemia with an 8; 14 and 14; 18 chromosome translocation. Surface marker studies of the cell line showed that the cells were positive for HLA-DR, CALLA (CD10), Bl (CD20) and B4 (CD19), but negative for Tl 1 (CD2). The fresh cells from peripheral blood of the patient had no surface immunoglobulins, whereas KHM-2B cells were positive for μ·λ type surface immunoglobulin. A cytogenetic analy… Show more

“…2b). This change is observed for both the L3͞Bcl-2 transfectant and KHM-2B, an unusual BL line carrying (in addition to the hallmark 8:14 myc-translocation) a 14:18 translocation juxtaposing bcl-2 to the IgH chain locus (18).…”

“…WILL2 was established from a patient with DLBCL showing surface IGλ and t(8;22)(q24;q11) in our laboratory . KHM10B and KHM2B were established from patients diagnosed with B‐cell type of acute lymphoblastic leukemia (FAB: L3) (purchased from JCRB, Osaka, Japan) . Both KHM10B and KHM2B are positive for surface IGλ.…”

“…In the patient sample and KMS-12PE, 4.5- and 1.5-kb mRNAs for the PRAD1/Cyclin D1 gene were detected (arrows). Lane 1: 1 µg of the total RNA from patient’s peripheral blood mononuclear cells; lane 2: 0.5 µg of the total RNA from KMS-12PE (a myeloma cell line with translocation 11;14) [26] as a positive control; lane 3: 5 µg of the total RNA from KHM-2B (a B-acute lymphoblastic leukemia cell line with translocations 8;14 and 14;18) [27] as a negative control. b Ethidium-bromide stained gel.…”
