“…, and L15 (Thermo Fisher Scientific), supplemented with 2 mM CaCl 2 , 2% FBS, and bovine insulin (5 μg/ml). CHO cells stably expressing an active form of H-Ras (CHO-Ras cells) were provided by Sanetake Shirahata (Kyushu University, Fukuoka, Japan), and CHO-Ras cells stably expressing mouse SIRPα, mouse CD47, or human SIRPα were provided by Nakayuki Honma (Kyowa Hakko Kirin, Tokyo, Japan) (58,59). CHO-Ras cells and their derivatives were cultured in α-modified minimum essential medium (Sigma-Aldrich) supplemented with 2 mM L-glutamine, 10 mM HEPES-NaOH (pH 7.4), and 10% FBS.…”

“…We previously showed that SIRP␣ is abundantly expressed in BMDCs [20]. We thus next examined the migration of BMDCs, which had been subcutaneously injected into the footpads, to the draining LNs.…”

“…In contrast, CD47 is expressed in a variety of hematopoietic cells including red blood cells and T cells [13][14][15]. We have recently shown that mice expressing a mutant form of SIRP␣ that lacks most of the cytoplasmic region are resistant to 2,4-dinitro-1-fluorobenzene (DNFB)-induced CHS as well as to experimental autoimmune encephalomyelitis (EAE) or collageninduced arthritis [20][21][22]. In addition, the production of IL-17 by T cells from SIRP␣ mutant mice exposed to the antigens responsible for the induction of these conditions was markedly impaired, suggesting that SIRP␣ expressed on the surface of DCs is essential for priming of naive T cells as well as for their Th17 differentiation during the development of CHS or EAE.…”

“…CHO cells stably expressing an active form of H-Ras (CHO-Ras cells) and those stably expressing mouse SIRPa/CD172a (CHO-Ras-SIRPa-ICR) were cultured in a-MEM medium (Sigma-Aldrich) supplemented with 10% FCS, 2 mM L-glutamine, 10 mM HEPES, 500 mg/ml Geneticin, and 500 mg/ml Zeocin, as previously described (27). COS-7 cells were maintained in DMEM supplemented with 10% heat-inactivated FCS, L-glutamine, and 0.1 mM nonessential amino acids at 37˚C with 5% CO 2 and transfected with a pCMV-SPORT6 expression vector encoding mouse CD63 Ag (Invitrogen Life Technologies) to generate a stable cell line expressing CD63 on the cell surface.…”
