The essential oil composition of Thymus vulgaris L. and Rosmarinus officinalis L. endemic to France were determined by GC and GC-MS. Oils were assessed for their cytotoxic, antioxidant and antimicrobial activity. 31 and 37 different compounds were identified representing 99.64% and 99.38% of the thyme and rosemary oils respectively, where oxygenated monoterpenes constituted the main chemical class. Thymol (41.33%) and 1.8-cineole (24.10%) were identified as the main constituents of T. vulgaris L. and R. officinalis L., respectively. Essential oils (EOs) of selected plant species were evaluated for their in vitro cytotoxicity against the human lung adenocarcinoma epithelial cell line (A549). Cytotoxicity was measured using MTT [3-(4,5-dimethylthiazol-2-yl)-2,5-diphynyltetrazolium bromide] colorimetric assay. Dose-dependent studies revealed IC 50 of 8.50 ± 0.01 µg/mL and 10.50 ± 0.01 µg/mL after 72 h on the A549 cells for R. officinalis L. and T. vulgaris L., respectively. Antioxidant activity was determined using a quantitative DPPH (1,1-diphenyl-2-picryl hydrazyl) assay. Thymus and rosemary EOs exhibited effective radical scavenging capacity with 50% inhibitory concentration (IC 50 ) of 437 ± 5.46 µg/mL and 189 ± 2.38 µg/mL respectively and therefore acts as a natural antioxidant agent. The antimicrobial activity of these species has also been studied against several foodborne pathogens and food isolated Salmonella spp. including S. enteritidis of significant importance. According to the results, T. vulgaris L. showed higher bactericidal effect than those from R. officinalis L. These results suggest that the essential oil from T. vulgaris L. and R. officinalis L. have potential to be used as a natural cytotoxic, antioxidant and antimicrobial agent in food processing.

Introduction:The world health organization recommends validating medicinal plants to ensure safety and efficacy. Lavandula is valued for the production of essential oil used in aromatherapy. Rosmarinus is known for its folk use as anti-oxidant and antidiabetic. This study aims to design monographs for Rosmarinus officinalis (Linn.) and Lavandula angustifolia (Linn.) cultivated, in conditions similar to their natural habitat, at Beirut Arab University botanical garden. Also screening the antioxidant activity of both plants, and assessing the antidiabetic effect of R. officinalis (Linn.). Methods: macroscopic and microscopic examination, hydrodistillation to obtain essential oils (EOs), thin layer chromatography, Gas chromatography-Mass spectrometry, and physiochemical properties of the EOs were determined. The antioxidant effect of the EOs is determined by DPPH assay. The antidiabetic effect of Rosemary EO is investigated using alloxan-induced diabetic mice. Results: the qualities of studied species are in agreement with the standards described in WHO monographs and in the British pharmacopeia. Composition of the EO produced from either Rosemary (REO) or Lavender (LEO) collected at different months showed differences. The EO of both showed significant antioxidant effects in DPPH assay (p<0.05). In vivo investigation of the antidiabetic effect of REO showed a significant antidiabetic effect in diabetic mouse model (p<0.05). Conclusion: both plant species and their EOs fulfilled most of standards specified by the British Pharmacopeia 2013. Rosemary collected in April (Spring) showed the best antioxidant activity. Lavender collected in September (Early Fall) showed the best antioxidant activity. REO revealed significant lowering in blood glucose level at the dose of 110 mg/kg.

RESUMENEl objetivo del presente trabajo fue evaluar el efecto de las microemulsiones de aceite esencial de romero (AER) y ár-bol de té (AET) sobre el eritrocito humano y microorganismos patógenos. Para ello, se elaboraron microemulsiones de AER y AET al 8.0% (v/v), 5.0% (v/v) y 2.5% (v/v). Las microemulsiones fueron probadas sobre el eritrocito humano para determinar el porcentaje de hemólisis, el porcentaje de inhibición de hemólisis y su actividad antibacterial contra E. coli O157:H7 y S. aureus. Las microemulsiones con AER no presentaron actividad hemolítica significativa, caso contrario con las microemulsiones de AET al 8.0% (≈70%) y 5.0% (33%) que presentaron mayor actividad hemolítica. Las microemulsiones de AER protegieron significativamente al eritrocito contra la presencia de radicales libres, en comparación con aquellas de AET (p< 0.05). Además, las emulsiones de AET al 8.0% mostraron efectos antibacterianos contra E. coli O157:H7 y S. aureus mientras que AER al 8.0% solo mostraron efecto contra E. coli O157:H7. La limitante del estudio fue que no utilizamos células nucleadas para establecer si los aceites esenciales dañan el material nuclear. Sin embargo, observamos que el tipo y la cantidad de aceite utilizado pueden tener implicaciones serias sobre la membrana eritrocitaria. Se concluye que las microemulsiones de AER presentaron mejor efecto protector eritrocitario, mientras que las microemulsiones de AET presentaron mejor actividad antibacterial contra las bacterias estudiadas, pero con mayor efecto tóxico sobre el eritrocito.
