The mushroom Hericium erinaceus has been used as a food and herbal medicine since ancient times in East Asia. It has been reported that H. erinaceus promotes nerve growth factor secretion in vitro and in vivo. Nerve growth factor is involved in maintaining and organizing cholinergic neurons in the central nervous system. These findings suggest that H. erinaceus may be appropriate for the prevention or treatment of dementia. In the present study, we examined the effects of H. erinaceus on amyloid β(25-35) peptide-induced learning and memory deficits in mice. Mice were administered 10 μg of amyloid β(25-35) peptide intracerebroventricularly on days 7 and 14, and fed a diet containing H. erinaceus over a 23-d experimental period. Memory and learning function was examined using behavioral pharmacological methods including the Y-maze test and the novelobject recognition test. The results revealed that H. erinaceus prevented impairments of spatial short-term and visual recognition memory induced by amyloid β(25-35) peptide. This finding indicates that H. erinaceus may be useful in the prevention of cognitive dysfunction.

Hericium erinaceus is a medicinal mushroom producing biologically active metabolite erinacin A. In this research we compared fungal growth and metabolite production in different reactor types and different substrates with added casein peptone and NaCl. Cultivation of fungal biomass was performed in glass jars and horizontal stirred tank reactor. The cultivation in reactor was carried out at 24°C and airflow 5 L/min. Periodical mixing of N = 80 rpm, was used. Solid state substrate based on mixture of husked millet and paddy millet was used. For simultaneous determination of erinacine A and ergosterol contents in a single sample new analytical method was developed. The amount of biomass obtained in glass jars averaged at 100 mg/g ± 12 mg/g, while in horizontal mixing bioreactor it easily raised up to 350 mg/g. The highest erinacin A concentration was detected in substrate with 0.56% NaCl and 3.4% casein peptone content.

Neurotrophic effects of simplified mastigophorene analogs on mesencephalic dopaminergic cells in primary culture

Mastigophorenes are dimeric isocuparane‐type sesquiterpenes of plant origin which have been found to exhibit neurotrophic properties. In this study, a simplified synthetically produced analog (1) and its dimethylether (2) were screened for potential neurotrophic effects on primary dopaminergic cell cultures in vitro. Dopaminergic cultures were prepared from embryonic mouse mesencephalon and 6‐day serum‐free cultures were exposed to low concentrations (0.05–5.0 μM) of these compounds for 6 days. Although differing by only two O‐methyl groups, the two substances differed in their effects on dopaminergic cells. 1 promoted growth of individual cells at low concentrations, whereas the more lipophilic 2 prevented cell death in culture and also clearly stimulated the growth of neuronal processes, although effects on growth of the total cell area were less pronounced. Effects on mitochondrial respiratory enzymes appear unlikely as 1 and 2 showed inhibitory effects only at concentrations 10–100‐fold higher than those used in cell culture. The results obtained show that synthetically produced substances analogous to neuron‐supportive mastigophorenes of plant origin can serve as neurotrophic substances. Drug Dev.
