Epithelia: lymphocyte interactions in the gut

Abstract: The mucosal immune system is governed by a unique set of rules and regulations. The local microenvironment dictates the necessity for these differences. The intestinal epithelial cell (IEC) sits at the interface between an antigen-rich lumen and a lymphocyte-rich lamina propria (LP). The cross talk that occurs between these compartments serves to maintain intestinal homeostasis. IECs have the capacity to talk to LP lymphocytes, activating populations of unique regulatory T cells. These cells have the capacity … Show more

“…This process confers specificity to CSR 38 , because the promoter upstream of each C H gene responds only to a specific set of signal-induced transcription factors 10 . Promoters upstream of C α genes, or I α promoters, become activated in response to transforming growth factor-β1 (TGFβ1) [39][40][41] , a cytokine that is secreted by many cell types, including various subsets of CD4 + T cells 42,43 . Together with CD40L, TGFβ1 is essential for the induction of T-cell-dependent IgA class switching 10 .…”

“…In the intestine, two major lymphocyte populations exist: the intraepithelial lymphocytes (IEL) which remain associated with the basolateral membrane of the IECs, and the LPL which localize to the subepithelial lamina propria and are in contact with IECs via basolateral projections through the semi-porous basement membrane 1,2 . Previous studies from our lab suggested that IECs function as antigen-presenting cells (APCs) and, as such, can promote regulatory T-cell responses in the mucosa [3][4][5][6][7][8] .…”

“…Food allergy to wheat is typically characterized by a T helper type 2 (Th2) lymphocytic inflammation [124][125][126][127][128] in genetically predisposed [125,129,130] individuals. The subsequent cascade of Th2 activation could either lead to an IgE mediated (immediate response with wheatspecific IgE antibodies) or a non IgE mediated allergy (chronic cellular inflammation with presence of lymphocytes and eosinophils) [124][125][126][127][128].…”
