Epilobium angustifolium extract demonstrates multiple effects on dermal fibroblasts in vitro and skin photo-protection in vivo

Abstract: Stress-induced fibroblast senescence is thought to contribute to skin aging. Ultraviolet light (UV) radiation is the most potent environmental risk factor in these processes. An Epilobium angustifolium (EA) extract was evaluated for its capacity to reverse the senescent response of normal human dermal fibroblasts (NHDF) in vitro and to exhibit skin photo-protection in vivo. The HPLC-UV-MS analysis of the EA preparation identified three major polyphenol groups: tannins (oenothein B), phenolic acids (gallic and … Show more

“…The spontaneously immortalized human keratinocyte cell line HaCaT (gift of Prof. Dr. N. Fusenig (Boukamp et al , 1988)) was grown in Dulbecco's modified Eagle's medium (DMEM, Sigma, St Louis, Missouri) supplemented with 10% fetal bovine serum (BFS, Invitrogen, Carlsbad, CA, USA), glutamine (0.29 mg/ml) (Sigma) and gentamycin (50 µg/ml) (Invitrogen) in 5% CO 2 at 37 °C, as described previously (Ruszova et al , 2013; Ruszova et al , 2008). …”

“…NHDF were obtained from the dermis by the digestion method [20]. Briefly, tissue specimens ($3 cm 2 ) were rinsed with sterile phosphate buffer saline (PBS) and cut into smaller pieces ($0.5 cm 2 ), and the subcutaneous fat and epidermis were removed.…”

“…Ruszova et al (2013) 73 showed, by in vitro assays, beneficial effect of Epilobium angustifolium extract, after exposure, through downregulation of expression of MMP-1 genes and MMP-3 (collagenases) and hyaluronidase-2. Considering keratinocytes cell lines it is possible to recognize the properties of antioxidants in reducing the formation of free radicals after UVB radiation or induced by hydrogen peroxide in cell cultures (HaCat).…”
