Enzyme-linked immunosorbent assay for diagnosis of brucellosis

Abstract: The performance of an ELISA for detection of total antibodies to Brucella spp. was compared with that of the Rose Bengal, standard agglutination and Coombs test in the diagnosis of brucellosis. Sera tested were from 208 patients from whom Brucella melitensis had been isolated, 177 patients with significant results in at least two conventional tests, and 107 patients with fever from whom no Brucella spp. had been isolated and in whom all conventional tests were negative. ELISA was the most sensitive test (97%),… Show more

“…According to recent studies by Smits et al (35), 11% of patients with brucellosis do not have detectable levels of specific IgM by immunoblot analysis. Similar results were found with enzyme immunoassay techniques (3,4,6,34). In these cases, seroconversion accompanied by a significant rise in antibody titer allows brucellosis to be diagnosed.…”

“…The most commonly used tests are the serum agglutination test (SAT), the Coombs anti-Brucella test, the Rose Bengal test, and complement fixation. During the last decade, radioimmunoassay (24,28) and enzyme immunoassay (6,21,34) tests have also been used. These present technical difficulties since they require skilled personnel and high-cost material.…”

“…In 13 of the 16 studies included in this review, standard RBT performance was compared to culture as the reference test [ 2 , 15 , 19 – 21 , 23 , 27 , 28 , 30 32 ], and in 3/16 studies [ 2 , 23 , 34 ], titrated RBT (involving serum dilutions made in phosphate-buffered saline and then tested with an equal volume of the RBT reagent) was compared to culture ( Table 1 ).…”

“…However, care should be taken not to vaccinate pregnant animals. 5) To ensure a sound population of Holstein cows within the HPI scheme, this ELISA test should be repeated annually since it is a more sensitive test compared to the Rose Bengal, standard agglutination and Coombs test in the diagnosis of brucellosis (Saz et al 1987). 6) Artificial insemination with healthy semen should be imperative for the Heifer International scheme to avoid contamination of these valuable animals from infected bulls.…”
