Enzyme immunoassay of mumps virus in cell culture with peroxidase-labelled virus specific monoclonal antibodies and its application for determination of antibodies

Abstract: SummaryMumps neutralizing monoclonal antibodies (MAs) were purified and labelled with horseradish peroxidase and used to detect virus-infected Vero cells, which were seeded as monolayers in wells of 96-well plates. This direct enzyme immunoassay (EIA) in cell culture proved to be a sensitive method for detection and titration of mumps virus and it may be useful for diagnostic purposes. The EIA is also suitable for the rapid determination of neutralizing antibodies. Neutralization of mumps virus by preincubatio… Show more

“…We used only selected cells derived from African green (vervet) monkeys because primary African green kidney cells are known to be less sensitive than primary rhesus kidney cells for isolating paramyxoviruses [-5, 9]. Consideration was also given to previous studies which had shown some promise with Vero cells [16,17,23,26,27,33], LLC-MK2 [12,21], A-549 cells [30,34], and others [29,31,32].…”

“…MAb. The production and characterization of mumpsspecific monoclonal antibody (MAb) UM 10B (immunoglobulin G2a) have been described previously in another paper (12). Horseradish peroxidase (HRP) was conjugated to unpurified (ascitic fluid) MAb UM 10B by using the periodate method (8).…”

“…For TGE infections, this technique using infected cells as a source of antigen has been used to detect total anti-TGEV antibodies (Kodama et al, 1980a) or IgA antibodies (Kodama et al, 1980b) in the serum of infected swine. For other viruses, it has been used for screening hybridomas (Russell et al, 1983), for diagnosis (Pauli et al, 1984;Groin and Bernard, 1985;Van Tiel et al, 1988) and for identification of epitopes on enveloped or naked viruses (Anderson et al, 1986;Boere et al, 1986;Oosterlaken et ai., 1989;Simkins et al, 1989). Recently, we described a quantitative fixed-cell immunoperoxidase technique for the study of surface viral antigens induced by TGEV (I"6 et al, 1991).…”
