Enucleated GV Oocytes as Recipients of Embryonic Nuclei in the G1, S, or G2 Stages of the Cell Cycle

Abstract: Universal recipients in the G2 phase of mitotic cell cycle (preactivated oocytes, zygotes, blastomeres) accept embryonic nuclei in all the stages of their cell cycle. To test if recipients in the G2 of meiotic cycle (immature oocytes) are universal recipients, mouse germinal vesicle (GV) oocytes were enucleated and reconstructed with blastomere nuclei in the G1, S, or G2 stages. Analysis of their maturation has shown that about 30% of the G1 nuclei and 60% of G2 nuclei allow for normal metaphase II (MII), both… Show more

“…In Xenopus, Gurdon et al ruptured the GV by a needle to promote the mixing of GV contents with the oocyte cytoplast prior to nuclear transfer and significantly increased the efficiency of nuclear reprogramming [29]. The transfer of mouse somatic cells into GV oocytes confirmed that GV material was essential for nuclear reprogramming [30]. Nuclear components extracted from the GVs have been demonstrated to be important in facilitating reprogramming of the introduced foreign nuclei in mammals [31].…”

“…The results imply that GV stage cytoplasts are incompetent to support cloned embryo development. An earlier study by Mohammed et al also concluded that GV stage oocytes were unsuitable as recipients for nuclear transfer [30]. It is thought that the reprogramming factor(s) responsible for embryonic stem cells reside in the nucleus [35,36].…”

“…Treatments of dysfunctions or improvements of physiological and reproductive performances were achieved through feed supplements Mohammed and Al-Hozab, 2020;Sheoran et al, 2018) and assisted reproductive techniques (Mohammed et al, 2010;Mohammed et al, 2012;Mohammed 2019). The continuous interests for the use of dietary supplements have grown to human and animals as well in order to improve their productive, reproductive and therapeutic performances (Gonzalez-Uarquin et al, 2020).…”
