Enhancement of Fe(III), Co(III), and Cr(VI) reduction at elevated temperatures and by a thermophilic bacterium

Abstract: An unusual thermophilic bacterium has been isolated from deep subsurface sediments and tested for its ability to enhance Fe (m), Co (m>, and Cr (Vi) reduction. Without the * bacterium, abiotic metal reduction was insignrfcant at temperams below 45OC but became a major process at 75OC. Addition of the bacterium enhanced the reduction of these metals up to fourfold. This study demonstrates abiotic and biotic metal reduction under organicrich thermic conditions and suggests that thermally and/or biologically enha… Show more

“…Cell enumeration was performed using fluorescence microscopy (Hobbie et al 1977). HClextractable Fe(II) was evaluated using ferrozine (Zhang et al 1996). Reduction of Co(III) to Co(II) was evaluated with a spectrophotometer by measuring absorbance at 535 nm (Zhang et al 1996).…”

“…Bacterial numbers were determined by directly counting cells, which were stained with 0.1% acridine orange, under an epifluorescence microscope (57). A 0.1-ml sample of each dilution was inoculated into 28-ml pressure tubes containing 10 ml of basal medium and 0.05% yeast extract, with ferric citrate (10 mM) as the electron acceptor and hydrogen as the primary electron donor under a H 2 -CO 2 gas headspace.…”

“…To examine the chemical conditions of metal reduction and mineral formation by the thermophilic Fe(III)-reducing bacteria, subsamples (1 ml) of bacterial cultures and abiotic controls were taken from the culture bottles at different times and measured for redox potential (E h ) and pH at room temperature in an anaerobic chamber (58). Fe(II) concentrations (0.5 N HCl, soluble) were determined by measuring the absorbance at 562 nm on a Hewlett-Packard model 8453 spectrophotometer by the ferrozine method with anaerobic water for sample dilution (50,57).…”
