Enhanced thebaine production inPapaver bracteatumcell suspension culture by combination of elicitation and precursor feeding

Abstract: In this study, the effect of methyl jasmonate (MJ) and ultrasound (US), individually and in combination with L-tyrosine, on the stimulation of thebaine production in Papaver bracteatum cell suspension cultures was studied. The addition of L-tyrosine did not significantly affect the cell biomass, but significantly increased the thebaine yield of cells compared with the control. The synergistic effects of MJ and L-tyrosine in the combined treatment of 100 μM MJ and 2 mM L-tyrosine increased the thebaine yield of… Show more

“…Likewise, MeJA + Phe produced an accumulation of phenols higher than the untreated samples (+ 56%) but lower than the MeJA + Tyr treatment. These data are in agreement with those reported for cell suspensions of other species such as Withania somnifera , Papaver bracteatum, Catharanthus roseus , and Vitis vinifera [26,28,29].…”

“…Therefore, the hemp cell suspensions were treated with MeJA + Phe or with MeJA + Tyr. Phe was chosen because it represents the substrate of PAL, the first committed enzyme of secondary metabolism, and Tyr was chosen since it is the precursor for thousands of specialized compounds [26,27]. Based on the amount of phenols that accumulated, the best combination proved to be MeJA + Tyr during 4 days of treatment, with an 82% phenol increase over the control cells.…”

“…The ultrasound treatment at 300 W for 5 min improved the conversion frequencies of protocorm like bodies (PLBs) to shoot in the Dendrobium officinale (Wei et al 2012). In Zare et al (2014) study, the combination of ultrasound (10 s) and L-tyrosine feeding (2 mM) significantly increased the production of thebaine in comparison to individual utilisation of 2 mM L-tyrosine and ultrasound (10 s). 1d-f) data revealed significant differences (P≤0.01) between different PGRs.…”

“…At the end of this period (42 days), several parameters such as the percentage of callus induction, callus morphology, callus fresh weights (CFW) and callus dry weights (CDW) were measured. Cell suspension culture was started according to protocol as descripted by Zare et al (13). Briefly, friable root callus (80-100 mg) transferred into a 50 mL Erlenmeyer flask containing 15 mL of liquid 3:4 MS medium filled with 3 mg L -1 2,4-D and 0.5 mg L -1 BAP.…”

“…In plant cells, one of the defense response is the accumulation or even production of secondary metabolites. These responses can be activated by different endogenous and exogenous molecules called elicitors (13). One of the important elicitors, which naturally exist in higher plants is methyl jasmonate (MJ).…”
