Enhanced Chemo‐Immunotherapy Strategy Utilizing Injectable Thermosensitive Hydrogel for The Treatment of Diffuse Peritoneal Metastasis in Advanced Colorectal Cancer

Meng Wang,
DanRong Hu,
Yun Yang
et al.

Abstract: Patients with colorectal cancer (CRC) and diffuse peritoneal metastasis (PM) are not eligible for surgical intervention. Thus, palliative treatment remains the standard of care in clinical practice. Systemic chemotherapy fails to cause drug accumulation at the lesion sites, while intraperitoneal chemotherapy (IPC) is limited by high clearance rates and associated complications. Given the poor prognosis, a customized OxP/R848@PLEL hydrogel delivery system has been devised to improve the clinical benefit of adva… Show more

“…It undergoes phase transition to form a drug-loaded gel depot for sustained drug release upon in vivo administration. Moreover, PLEL hydrogels effectively prolong drug retention time [ 25 , 26 ]. Nevertheless, the poor solubility of small-molecule drugs makes it difficult to achieve a uniform dispersion within hydrogels.…”

“…Furthermore, activated CD8 + T cells secrete IFN-γ and TNF-α, initiating a cascade of inhibitory immune responses against the tumor. Conversely, TGF-β hinders pro-inflammatory cells and Th cells while promoting Treg cells ( 35 , 36 ). Illustrated in Figures 3E–I , is an escalation in IFN-γ and TNF-α secretion post-combination therapy, indicating a heightened immune response of CD8 + T cells.…”
