Enhanced cell disruption strategy in the release of recombinant hepatitis B surface antigen from Pichia pastoris using response surface methodology

Abstract: BackgroundCell disruption strategies by high pressure homogenizer for the release of recombinant Hepatitis B surface antigen (HBsAg) from Pichia pastoris expression cells were optimized using response surface methodology (RSM) based on the central composite design (CCD). The factors studied include number of passes, biomass concentration and pulse pressure. Polynomial models were used to correlate the above mentioned factors to project the cell disruption capability and specific protein release of HBsAg from P… Show more

“…The advancement of molecular biology, PCR technique has been broadly used to amplify the desired gene and had been proven as one of the most effective molecular diagnoses (Sadiq et al, 2017;Chan et al, 2007). An amplified gene products can be used as important genetic materials for further genetic analysis and gene expression studies (Razis et al, 2006;Tam et al, 2012;Ismail et al, 2012) functions. Furthermore, PCR could be utilized to produce complete genome sequence by closing the gaps in genome scaffold thus providing a valuable information regarding any organism (Balakrishnan et al, 2015) PCR technique is well known to produce quick confirmatory test for ORFV.…”

“…The mechanical methods including bead mill and high‐pressure homogenizer are based on solid shears and liquid shears, respectively, and they are widely used in commercial scale . The robustness of these techniques on cell disruption is well proven; however, since these techniques are non‐specific in cell disruption, they make the subsequent clarification and purification processes more challenging .…”
