Enhanced biostability and cellular uptake of zinc oxide nanocrystals shielded with a phospholipid bilayer

Abstract: The surface chemistry and charge of zinc oxide nanocrystals influence their behaviour in biological fluids. A novel lipid bilayer assembly is developed to shield ZnO nanocrystals improving their stability and cell internalization.

“…The reaction path is based on the hydrolysis of the zinc precursors (zinc acetate dihydrate) due to the presence of sodium hydroxide as the base in methanol. The as-synthesized ZnO were then functionalized with amino-propyl groups with a post-grafting approach using 3-(AminoPropyl)-TriEthoxySilane (APTES) at 10 mol% with respect to the molar amount of ZnO, as in Dumontel et al (2017) and Garino et al (2019a).…”

“…In previous researches, different methods were followed to decrease agglomerations, and improve dispersion, stability, and biocompatibility of the nanomaterials [12,13], such as an organic coating or addition of a capping agent (polyethylene glycol (PEG), l-dopa, poly-n-vinyl-2-pyrrolidone (PVP) and others [14,15]. However, the cytotoxic effects of capping agents or coatings have also been reported [16].…”

“…ZnO NPs (15 nm) decorated with both (1,2-dioleoyl-sn-glycero-3-phosphocholine) (DOPC) and amine-propyl chains exhibited a positive charge in water. However, DOPC-ZnO NPs were internalized at a significantly higher level than were NH 2 -ZnO NPs in HeLa cells, a distinction that may be due to the difference in positive charge (Dumontel et al, 2017). Moreover, the uptake level that is affected by surface charge may vary with cell type.…”
